COX-2与MMP-14在肾癌组织中的表达水平及临床意义
Expression Levels of COX-2 and MMP-14 in Renal Cell Carcinoma and Clinical Significance
【摘要】 目的研究环氧合酶-2(COX-2)和基质金属蛋白酶-14(MMP-14)在肾癌组织中的表达及其与肾癌浸润转移的关系。方法运用免疫组织化学SP法,分别检测45例肾癌和10例正常肾组织中COX-2和MMP-14的表达情况,结合临床病理参数进行分析。结果 COX-2在肾癌组织中的阳性表达率为55.6%(25/45),高于正常肾组织10%(P<0.05),COX-2的表达与肾癌淋巴结转移、浸润程度、临床分期和分化程度密切相关(P<0.05),与患者年龄和性别无关(P>0.05)。MMP-14在肾癌组织中的阳性表达率为66.7%(30/45),显著高于正常肾组织0%(P<0.05),MMP-14的表达与肾癌淋巴结转移、分化程度及浸润程度相关(P<0.05),与临床分期、患者性别和年龄无关(P>0.05)。COX-2与MMP-14在肾癌组织中的表达密切相关(P<0.05)。结论 COX-2和MMP-14在肾癌中高表达可能是促进肾癌浸润和转移的因素之一,两者可能是反映肾癌生物学预后的指标,并为探讨临床治疗肾癌的方法提供有力的依据。
【Abstract】 Objective To study the expressions of cyclooxygenase- 2( COX-2) and matrix metalloproteinase- 14( MMP- 14) in renal cell carcinoma( RCC) and the relationship of expressions with invasion and metastasis. Methods The expressions of COX-2 and MMP- 14 in 45 cases of renal cell carcinoma and 10 cases of normal renal tissues were detected by immunohistochemistry SP to analyze the relationship combined with clinical pathological parameters. Results The positive expression rate of C0X- 2 in RCC was 55. 6%( 25 /45),which was 10% higher than that in the normal renal tissue( P < 0. 05). The expressions of COX-2 were closely related to lymph node metastasis and invasion,clinical staging and the degree of differentiation( P < 0. 05),but not associated with patients’ age and sex( P > 0. 05). The positive expression rate of MMP- 14 in RCC was 66. 7%( 30 /45),which was significantly higher than that in normal renal tissue( P < 0. 05). The expression of MMP- 14 was related to lymph node metastasis,differentiation and invasion( P < 0. 05),but not associated with clinical staging and patients’ age and sex( P > 0. 05). Close correlation of C0X- 2 and MMP- 14 was found in RCC( P < 0. 05). Conclusion The high expression of COX-2 and MMP- 14 in RCC may be a cause to invasion and metastasis of RCC. COX-2 and MMP- 14 can be used as prognostic indicators of biology of renal cancer and provide powerful evidence for the clinical treatment of renal cancer.
