姜黄素对人肝癌HepG2细胞Notch1和NF-κB表达的影响
The effect of Curcumin on the expression of Notch1 and NF-κB in human hepatocellular carcinoma HepG2 cells
【摘要】 目的研究姜黄素作用于人肝癌HepG2细胞后Notch1、NF-κB的表达,探讨姜黄素抑制肝癌细胞增殖的机制。方法将肝癌细胞株HepG2培养至对数生长期,分组加入不同浓度(20、40、80μmol/L)的姜黄素干预后继续培养24 h,并设立空白对照组。用MTT法观察姜黄素对体外培养的肝癌细胞生长的抑制作用;q RT-PCR法检测各组Notch1m RNA表达量,Western blot法检测各组Notch1、NF-κB蛋白的表达水平。结果 MTT比色法测定显示,姜黄素对肝癌细胞HepG2的抑制作用呈剂量依赖性,对照组及不同浓度姜黄素作用组的抑制率分别为0%,25.36%±2.54%,44.64%±2.98%,74.94%±3.62%。q RT-PCR及Western blot法提示,与对照组相比,姜黄素作用组的Notch1及NF-κB的表达量均下降且呈剂量依赖性。结论姜黄素抑制肝癌细胞HepG2的增殖,其机制可能与其下调Notch1、NF-κB的表达有关。
【Abstract】 Objective This study was to investigate the expression of Notch1 and NF-κB in human hepatocellular carcinoma HepG2 treated by curcumin, and to explore the possible mechanism of inhibiting proliferation of liver cancer HepG2 cells.Methods HepG2 cells were cultured to exponential phase and divided into different groups in which curcumin of different concentrations(20,40,80μmol/L)was added for 24 hours and the group without adding curcumin was set up as the control.MTT assay was used to detect cell proliferation in different groups. The level of Notch1 m RNA was determined by q RT-PCR.The protein expression of Notch1 and NF-κB were detected by Western blot. Results Compared with the control group, MTT showed that curcumin could inhibit the proliferation of liver cancer HepG2 cell in a dose-dependent manner. It has shown that cellular proliferation inhibition rate were respectively 0%,(25.36 ±2.54)%,(44.64 ±2.98)%,(74.94 ±3.62)% in the control group and groups with curcumin concentration of 20,40,80μmol/L. q RT-PCR and Western blot results showed that in comparison to the control group, the expression of Notch1 and NF-κB both decreased in a dose-dependent manner.Conclusion Curcumin could suppress the growth of HepG2 cells obviously. The mechanism might be related to the down-regulation of Notch1 and NF-κB expression.
