21天不完全睡眠剥夺对雄性大鼠脂代谢的影响

【摘要】 【目的】观察不完全睡眠剥夺对雄性大鼠脂代谢的影响,并分析其可能的机制。【方法】将36只健康的3月龄雄性Wistar大鼠随机分为3组(12只/组),其中2组利用"小平台水环境法"建立大鼠睡眠剥夺模型(剥夺18 h组,剥夺22 h组),另一组正常环境进行饲养(对照组)。持续干预21 d后,股动脉放血处死大鼠,分离血清。测定总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、极低密度白蛋白(VLDL)、载脂蛋白-A1(Apo-A1)、载脂蛋白B(Apo-B)的含量,取出大鼠肝脏组织并对其肝脏进行称重和计算肝脏系数。【结果】剥夺18 h组和剥夺22 h组大鼠的体质量和肝脏重量均低于对照组(P<0.01)。剥夺22 h组大鼠血清中TC、HDL-C、LDL-C的含量高于对照组(P<0.01),VLDL和TG含量低于对照组(P<0.05);Apo-A、Apo-B的含量在各组之间无明显变化。【结论】长期不完全睡眠剥夺能够引起雄性大鼠脂代谢紊乱。

【Abstract】 【Objective】To observe the effects of long-term partial sleep deprivation(SD) on lipid metabolism of male rates and analyze its possible mechanisms.【Methods】Thirty-six male Wistar rats were randomly and equally divided into three groups: SD 1 group(sleep deprivation for 18 hours a day group), SD 2 group(sleep deprivation for 22 hours a day group), control group(cage control). The SD model was established in a "small platform water environment",and rats in the control group were raised in normal conditions. After21-day intervention, all rats were sacrificed by femoral artery bleeding, the serum was separated and the level of Tc, TG, HDL-C, LDLC, VLDL, apo-A1 and apo-B in serum were determined. Their livers were removed and weighed.【Results】After the sleep deprivation,the weights of Wistar rats and their livers in SD 1 group and SD 2 group were lower than those in the control group(P<0.01). The levels of TC、HDL-C、LDL-C in SD2 group were significantly higher than those in the control group(P<0.01). Compared with the control group,the concentrations of VLDL and TG in SD2 group were lower(P<0.05). The contents of apo-B in all groups showed no significant changes.【Conclusion】Long-term partial sleep deprivation can cause lipid metabolic disorder in male rates.
