Caspase-8在维甲酸致小鼠囊胚细胞凋亡中的作用
Effects of Caspase-8 on Retinoic Acid-induced Apoptosis in Mouse Blastocys
【摘要】 目的探讨半胱氨酸一天冬氨酸蛋白酶(Caspase)家族中的Caspase-8对维甲酸致小鼠囊胚细胞凋亡的影响。方法小鼠囊胚随机分为空白组、低浓度组(1μmol/l RA)和高浓度组(10μmol/l RA),分别于M199培养基中培养24h。用免疫组化S-P法观察囊胚细胞中Caspase-8蛋白表达。结果高浓度维甲酸可显著诱导囊胚细胞凋亡,上调囊胚细胞中Caspase-8蛋白表达水平。结论维甲酸对胚胎的致畸效应可能与其诱导细胞过度凋亡有关。
【Abstract】 Objective To observe the effects of Caspase-8 on retinoic acid-induced apoptosis in mouse blastocys in vitro. Methods Mouse 3.5 d blastocysts were exposed for 24 h to doses of 0μmol/l, 1μmol/l and 10μmol/l retinoic acid, Fixed blastocysts and observed for their expression of Caspase-8 proteins by immunohistochemical S-P staining. Results RA can induce apoptosis of mouse blastocysts and up-regulate the expression of Caspase-8 protein in mouse blastocysts. Conclusion The teratogenic effect of retinoic acid on embryos may be related to the induction of excessive apoptosis.
