兔骨质疏松模型椎体成形术中骨水泥对邻近椎间盘的影响
Effect of Bone Cement in Percutaneous Vertebroplasty on Adjacent Intervertebral Disc in Osteoporosis Model of Rabbit
【摘要】 目的讨论兔骨质疏松模型椎体成形术中骨水泥对邻近椎间盘的影响。方法 5月龄新西兰雌性大白兔18只,随机分为三组:C组、T0组、T1组。经去势联合糖皮质激素注射造成骨质疏松模型后,对C组腰5椎体仅完成穿刺假手术,对T0组腰5椎体穿刺后注入碘海醇,对T1组腰5椎体穿刺后注入骨水泥,术后继续饲养12周。术后12周取腰4-5椎间盘,行TUNEL染色了解细胞凋亡率,行real-time PCR检测白介素1、Ⅱ型胶原蛋白、金属基质蛋白酶7m RNA相对表达量,各组数据进行比较并行统计学分析。结果 T1组凋亡细胞率同C组、T0组比较具有统计学差异;T1组白介素1、金属基质蛋白酶7m RNA相对表达量同C组、T0组比较明显升高并具有统计学差异;T1组Ⅱ型胶原蛋白mRNA相对表达量同C组、T0组比较明显降低并具有统计学差异。结论兔骨质疏松模型实施椎体成形术后,椎体内骨水泥确可对邻近椎间盘造成影响。
【Abstract】 Objective To determine the effect of bone cement in percutaneous vertebroplasty on adjacent intervertebral disc in osteoporosis model of rabbit. Methods Eighteen 5-months old New Zealand rabbits were randomly assigned to three groups:C,T0 and T1 groups.After establishing the osteoporosis model with the methord of ovariectomy combined with glucocorticoid injection,the fifth lumbar vertebras of C group were undergo sham operation of puncture,the fifth lumber vertebras of T0 group were injected with iohexol after puncture,the fifth lumber vertebras of T0 group were injected with bone cement after puncture,all three groups were raised for 12 weeks after operation.After 12 weeks L4-5 Intervertebral discs were taken out,the discs were done by TUNEL technique to determine the cell apoptosis rate,and the discs were done real-time PCR test to determine the relative expression of interleukin-1,type II collagen and matrix metalloproteinase-7.Data of each group were compared and done the statistical analysis. Results The cell apoptosis rate in T1 group was statistically significant compared with that in C and T0 group;The relative expression of Interleukin-1 and matrix metalloproteinase-7 in T1 group were higher than those in C and T0 group and had statistical significance; The relative expression of type II collagen in T1 group were higher than that in C and T0 group and had statistical significance. Conclusion After percutaneous vertebroplasty in osteoporosis model of rabbit,bone cement in the vertebra could influnce on the adjacent intervertebral disc.
