T-2毒素以及与DON联合作用对大鼠肝微粒体中CYP2E1含量及活性的影响
Effect of T-2 Toxin and DON Koinonia for CYP2E1 Content and Activity in Rat’s Hepatomicrosome
【摘要】 目的观察T-2毒素(T-2 toxin)以及与脱氧雪腐镰刀菌烯醇(deoxynivalenol,DON)毒素对大鼠肝微粒体中CYP2E1的影响,探讨其致病机理及联合毒素损伤效应。方法 30只四周龄雄性Wistar大鼠,按照完全随机化的方法分为三组:空白组、T-2毒素组、"T-2+DON"组(每组10只),染毒方法为模拟自然进食方式经口摄入掺毒饲料,T-2毒素含量为150μg/kg;联合组为T-2毒素100μg/kg,DON毒素220μg/kg。实验周期30日。处死,取肝脏,制备肝微粒体,ELISA测定大鼠肝微粒体CYP2E1的含量及活性,观察T-2毒素及与DON联合对肝脏的毒理作用。结果染毒30天后,空白对照组CYP2E1含量为(3.795±0.243)(ng/ml),T-2组为(2.342±0.137)(ng/ml),联合组为(2.309±0.137)(ng/ml),经单因素方差分析P<0.01,染毒组CYP2E1含量下降。空白对照组CYP2E1活性为(3.243±0.016)(u/ml),T-2组为(3.317±0.038)(u/ml),联合染毒组为(3.4±0.04)(u/ml),经单因素方差分析,染毒组的CYP2E1活性明显降低(P<0.01),T-2染毒组与联合染毒组(T-2+DON)之间肝微粒体中CYP2E1的活性差异无统计学意义(P>0.05)。结论 T-2毒素可致CYP2E1含量与活性降低,且T-2毒素和DON毒素联合作用更明显,这可能是机体T-2毒素和DON毒素中毒的机制之一。
【Abstract】 Objective to observe effect of T-2 toxin and deoxynivalenol(DON) toxin for CYP2E1 in rat’s hepatomicrosome,and explore pathogenic mechanism and combined toxin damage effect Methods divide 30 cases four-week old male Wistar rats according to completely randomized method into three groups:control group,T-2 toxin group,"T-2 + DON"group(n-10 for each),exposure method was simulating natural way of eating,intaking poison feed by oral,T-2 toxin content was150μg/kg;combination group was T-2 toxin 100μg/kg,DON toxin was 220μg/kg.Experimental cycle was 30 days.After killing them,prepare hepatomicrosome through liver,detect content and activity of CYP2E1 in rat’s hepatomicrosome by ELISA.Observe toxic effect of combined T-2 toxin and DON for liver.Results after 30 days of exposure,CYP2E1 content of blank control group was(3.795± 0.243)(ng/ml),that of T-2 group was(2.342±0.137)(ng/ml),and combination group was(2.309 ± 0.137)(ng/ml).Single factor variance analysis showed P<0.01,CYP2E1 content of exposure group decreased.CYP2E1 activity of blank control group was(3.243 ± 0.016)(U/ml),that of T-2 group was(3.317±0.038)(U/ml),and combined group was(3.4±0.04)(U/ml).single factor variance analysis showed CYP2E1 activity of exposure group decreased statistically(P<0.01),and difference of CYP2E1 activity in hepatomicrosome between T-2 group and combined group(T-2+DON) showed no statistical significance(P>0.05).Conclusion T-2 toxin can reduce CYP2E1 content and activity,and combined the effect of T-2 toxin and DON toxin is more obvious,which may be one of mechanisms of T-2 toxin and DON toxin poisoning
