浸润性CD11b+CD15+粒细胞在胃癌微环境中免疫抑制功能研究
Study on the immunosuppressive function of CD11b+CD15+ cells in microenvironment of human gastric cancer
【摘要】 目的探讨浸润性CD11b+CD15+粒细胞在胃癌微环境中免疫抑制功能。方法选取2011年2月至2015年2月44例手术标本作为研究对象,以CD11b+CD15+作为表面标志物,分离胃癌和癌旁正常组织浸润粒细胞,检测其比例和表型情况,并与胃癌临床特征进行比较。结果胃癌组织中性粒细胞含量显著高于癌旁正常组织,而IL-2、IFN-γ、颗粒酶B则显著低于癌旁正常组织,两者比较,差异有统计学意义(P<0.05);IL-2、IFN-γ含量,对照组最高,ARG-1阻断组其次,IL-10诱导组最低,三组比较,差异有统计学意义(P<0.05);CD+15粒细胞高浸润组和低浸润组在性别、年龄、肿瘤大小、胃壁浸润深度上比较,差异无统计学意义(P>0.05),而在TNM分期、淋巴结转移上比较,差异有统计学意义(P<0.05)。结论胃癌组织浸润性CD11b+CD15+粒细胞显著高于癌旁组织,胃癌组织浸润粒细胞依赖精氨酸酶Ⅰ从而发挥免疫抑制作用。
【Abstract】 Objective To investigate the immunosuppressive function of CD11b+CD15+ cells in gastric cancer microenvironment. Methods From Feb. 2011 to Feb. 2015,44 cases of surgical specimens were selected as study objects,CD11b+CD15+ was as a surface marker,gastric cancer and granulocyte infiltration were separated,the ratio of phenotype was detected,meanwhile,the clinical features of gastric cancer were compared among groups. Results The neutrophils of gastric cancer was significantly higher than that of adjacent tissues,while IL-2,IFN-γ,Granzyme B were significantly lower in gastric cancer than those in adjacent tissues( P < 0. 05). The levels of IL-2,IFN-γ were highest in the control group( control group > ARG-1 blocked group > IL-10 group),there was significant difference among three groups( P <0. 05). There was no significant difference of gender,age,tumor size,invasion depth between CD15+ granulocyte infiltration high group and low group( P > 0. 05). However,the results were opposite in TNM stage and lymph node metastasis(P < 0. 05). Conclusion The infiltration of CD11b+CD15+ granulocytes in gastric cancer tissues is significantly higher than that in the peripheral tissues,and the infiltration of granulocytes in gastric cancer is dependent on the expression of arginine.
