右旋雷贝拉唑钠对反流性食管炎模型大鼠炎性因子及血管活性肠肽的影响

【摘要】 目的探讨右旋雷贝拉唑钠对反流性食管炎模型大鼠炎性因子及血管活性肠肽(vasoactive intestinal peptide,VIP)的影响。方法选择60只SPF级大鼠为研究对象,随机分为空白对照组、模型组、雷贝拉唑钠组(4 mg/kg)、小剂量右旋雷贝拉唑钠组(2 mg/kg)和大剂量右旋雷贝拉唑钠组(4 mg/kg),空白组不作任何处理,其余4组建立大鼠反流性食管炎模型,观察各组大鼠食管炎指数、食管炎抑制率、炎性因子及VIP表达水平。结果大剂量右旋雷贝拉唑钠组食管炎指数明显低于雷贝拉唑钠组,抑制率明显高于雷贝拉唑钠组(t=3.725,7.595,P<0.05),一氧化氮(NO)、白细胞介素-6(IL-6)水平均明显低于雷贝拉唑钠组(t=2.054,6.386,P<0.05),VIP明显低于雷贝拉唑钠组,P物质(SP)明显高于雷贝拉唑钠组(t=2.826,3.162,P<0.05);小剂量右旋雷贝拉唑钠与雷贝拉唑钠食管炎指数等差异均无统计学意义(P>0.05)。结论右旋雷贝拉唑钠有通过降低炎性因子及VIP的表达,提高兴奋性多肽类神经递质的表达来发挥治疗反流性食管炎的作用,相同剂量条件下,右旋雷贝拉唑钠作用效果强于雷贝拉唑钠。

【Abstract】 Objective To investigate effect of dexrabepraxole sodoum on inflammatory factor and vasoactive intestinal peptide of reflux esophagitis model rats. Methods Sixty SPF rats were randomly divided into blank control group,model group,rabeprazole sodium group( 4 mg/kg),low-dose dexrabepraxole sodoum group( 2 mg/kg),high-dose dexrabepraxole sodoum group( 4 mg/kg). Blank control group was no any treatment,rats in other four groups were established reflux esophagitis rats model,then esophagitis index,esophagitis inhibition rate,inflammatory cytokines and vasoactive intestinal peptide expression level were compared. Results Esophagitis index in high-dose dexrabepra xole sodoum group was significantly lower than that in rabeprazole sodium group,inhibition rate was significantly higher than that in the rabeprazole sodium group( t = 3. 725,7. 595,P < 0. 05),NO,IL-6 levels were significantly lower than those in rabeprazole sodium group( t = 2. 054,6. 386,P < 0. 05),VIP was significantly lower than that in rabeprazole sodium group,SP was significantly higher than that in rabeprazole sodium group( t = 2. 826,3. 162,P < 0. 05); there were no statistical differences of esophagitis index and so on between low-dose dexrabepraxole sodoum group and rabeprazole sodium group( P > 0. 05). Conclusion Dexrabepraxole sodoum can reduce inflammatory cytokines and vasoactive intestinal peptide,improve the excitatory expressed peptide neurotransmitters more to play the role of the treatment of reflux esophagitis,under the condition of the same dose,the effect of dexrabepraxole sodoum is better than rabeprazole sodium.
