转染BRMS1基因对人类胰腺癌细胞在裸鼠体内肿瘤转移能力影响的研究
Study on BRMS1 gene transfection of human pancreatic cancer cells in nude mice effects of tumor metastasis
【摘要】 目的建立人类胰腺癌裸鼠皮下移植瘤模型,研究BRMS1基因对人类胰腺癌细胞在裸鼠体内肿瘤转移能力的影响,寻找胰腺癌基因治疗的有效模式。方法利用脂质体转染技术将构建的p EGFP-C1-BRMS1重组载体、p EGFP-C1空载体稳定转染入人类胰腺癌细胞株PANC-1中,接种三组(转染组、空载体组、未转染组)细胞于裸鼠背部皮下,5周后处死裸鼠,取肿瘤、淋巴结及肺组织进行组织病理学检查。结果将p EGFP-C1-BRMS1重组载体、p EGFP-C1空载体转染入人类胰腺癌细胞株PANC-1,并建立了人类胰腺癌裸鼠皮下移植瘤模型,三组裸鼠肿瘤大小无明显差异,但转染组裸鼠淋巴结转移率明显低于空载体组和未转染组。结论 BRMS1基因不影响人类胰腺癌细胞裸鼠皮下移植瘤的生长,但可以抑制人类胰腺癌细胞裸鼠皮下移植瘤的局部淋巴转移。
【Abstract】 Objective:The establishment of human pancreatic cancer nude mice model to study the BRMS1 gene in human pancreatic cancer cells in nude mice tumor metastasis ability to find an effective model of pancreatic cancer gene therapy.Methods:Liposome transfection technique using the constructed p EGFP-C1-BRMS1 recombinant vector,p EGFP-C1 empty vector stably transfected into human pancreatic cancer cell line PANC-1,the inoculated three groups(transfected group,empty vector group,not transfected group) cells subcutaneously in nude mice,mice were killed after 5 weeks,the tumor,lymph node and lung tissue for histopathological examination.Results:The p EGFP-C1-BRMS1 recombinant vector,p EGFP-C1 empty vector transfected into human pancreatic cancer cell line PANC-1,and established human pancreatic cancer xenograft model in nude mice,three groups no significant difference in tumor size in nude mice However,lymph node metastasis in nude mice transfected group was significantly lower than the empty vector group and untransfected group.Conclusion:BRMS1 gene in human pancreatic cancer cells without affecting tumor growth in nude mice subcutaneously,but can inhibit human pancreatic cancer cells transplanted into nude mice of the local lymph node metastasis.
