低剂量CoCl2预处理降低耳蜗毛细胞和螺旋神经元的顺铂毒性

【摘要】 目的研究低剂量氯化钴预处理对顺铂导致的耳蜗毛细胞和螺旋神经节损伤的保护作用。方法体外培养小鼠耳蜗基底膜,随机分为4组:正常组、100μM氯化钴处理组、200μM顺铂处理组、100μM氯化钴预处理加200μM顺铂干预组,通过免疫荧光染色观察细胞状态并计数,通过蛋白质印记法检测各组Caspase 3、HIF-1α、NF-kB表达量。结果氯化钴处理组与正常组之间无明显差异,顺铂处理组较正常组毛细胞和螺旋神经元损伤严重,氯化钴预处理加顺铂作用组毛细胞和螺旋神经元存活数目分别是顺铂处理组的2.81、1.40倍。氯化钴预处理组和顺铂组Caspase 3表达量分别比正常组高2.50、3.91倍。结论氯化钴预处理对减轻顺铂对毛细胞和螺旋神经元的氧化应激损伤有明显作用

【Abstract】 Objective To sduty the protection of low doses of CoCl2 pretreatment in cochlear hair cells and spiral ganglions which are injured by cisplatin.Methods We culture the cochlear basement membrane in vitro and divide them into four groups randomly:normal group,100μM CoCl2 treatment group,200μM cisplatin treatment group,100μM CoCl2 and pretreatment plus 200μM cisplatin intervention group.We observe the cell status and count with immunofluorescence staining,detect the expression quantity of Caspase 3,HIF-1α and NF-kB with Western blot.Results The CoCl2 treatment group is the same as the normal group,While the cisplatin treatment group has extremely injured hair cells and spiral ganglions compared to the normal group.The survival cell number of hair cell and spiral ganglions in CoCl2 pretreatment group is 2.81,1.4 times as many as the cisplatin treatment group.The western blot results indicate the expression quantity of CoCl2 pretreatment group and the cisplatin treatment group is 2.5,3.91 times as many as the normal group.Conclusion CoCl2 pretreatment can reduce the oxidative stress-mediated injury caused by cisplatin on hair cells and spiral ganglions.
