肾衰Ⅲ号方对阿霉素诱导损伤足细胞nephrin、desmin表达的影响
Effects of Shenshuai Ⅲ Mixture(肾衰Ⅲ号方) on Expressions of Nephrin and Desmin on Adriamycin-damaged Podocytes
【摘要】 目的:探讨肾衰Ⅲ号方对阿霉素诱导的足细胞损伤的保护作用。方法:分离大鼠肾组织足细胞以阿霉素0.5 mg/L培养24 h复制足细胞损伤模型,再分别以低、中、高剂量肾衰Ⅲ号含药血清(3.8%、7.5%、15%)干预24 h后,MTT检测细胞增殖活性,分别检测各组细胞nephrin、desmin mRNA(RT-PCR法)和蛋白(Western Blot法)表达。结果:MTT结果显示7.5%肾衰Ⅲ号含药血清干预24 h为效果最佳。RT-PCR结果显示,与正常对照组比较,模型组nephrin mRNA表达显著降低(P<0.01)、desmin mRNA表达显著增高(P<0.01);与模型组比较,高剂量组nephrin mRNA表达显著增高(P<0.05)、desmin mRNA表达显著降低(P<0.05)。Western Blot结果显示,与正常对照组比较,模型组nephrin蛋白表达显著降低(P<0.01),desmin蛋白表达显著增高(P<0.01);与模型组比较,中、高剂量组nephrin蛋白表达显著增高(P<0.01)、desmin蛋白表达显著降低(P<0.01)。结论:肾衰Ⅲ号含药血清能减轻阿霉素引起的原代培养的足细胞损伤。
【Abstract】 Objective: To investigate the protective effect of the Shenshuai Ⅲ Mixture on podocyte damaged by Adriamycin in vitro. Methods: The podocyte was isolated from rat renal tissue,and the model of podocyte damaged induced by 0. 5 mg·L-1adriamycin,then intervented by low,medium,high dose of Shenshuai Ⅲ Mixture containing serum( 3. 8%,7. 5%,15%),after 24 h,the expression of nephrin and desmin in each group was detected by RT-PCR and Western Blot. Results: MTT result showed that optimum observation condition was7. 5% containing serum of Shenshuai Ⅲ Mixture intevented 24 h. RT-PCR results showed that compared with control group,the nephrin mRNA expression of model group reduced significantly( P<0. 01),and desmin mRNA increased( P < 0. 01). Compared with model,nephrin mRNA of high dose of Shenshuai Ⅲ Mixture group increased( P< 0. 05),and desmin decreased( P < 0. 05). Western blot results showed that compared with control group,the nephrin of model group reduced significantly( P<0. 01),and desmin increased( P<0. 01). Compared with model,nephrin of medium dose and high dose group increased( P<0. 01),and desmin decreased( P<0. 01). Conclusion: Shenshuai Ⅲ Mixture could reduce podocyte damaged by Adriamycin.
