AC、PKA基因水平表达变化对心气虚型冠心病的相关性研究

【摘要】 目的探讨AC、PKA基因水平表达变化与心气虚型冠心病发病的相关性,以阐述心气虚型冠心病发病的微观机制。方法采用"冠状动脉结扎加注射左旋硝基精氨酸"法制备心气虚型冠心病大鼠模型,并将大鼠分为空白组、模型组、西药盐酸地尔硫卓组(3mg/kg)、益气活血方低、高、中剂量中药组(14.4,43.2,28.8g/kg),每组8只。造模成功后西药组用盐酸地尔硫卓干预,中药组用不同剂量的益气活血方干预,于造模45天后取心肌组织用PCR技术检测AC、PKA含量以观察基因水平表达变化。结果与模型组相比,中药组AC、PKA浓度均明显降低(P<0.01);与西药组比较高剂量中药组AC、PKA浓度均明显下降(P<0.01)。结论 AC、PKA基因水平表达变化与心气虚型冠心病存在相关性,AC、PKA基因水平表达变化在一定程度上揭示了心气虚型冠心病发病的微观机制。

【Abstract】 Objective The objective is to study the relativity between the expression of AC and PKA gene levels and the incidence of coronary heart disease insufficiency of the heart-Qi.In order to reveal the mechanism of pathogenesis of heart-Qi deficiency type of coronary heart disease.Methods A rat model of the heart-Qi deficiency type of coronary heart disease was established by coronary artery ligation and injection of L-arginine,The rats were divided into control group,model group,western medicine group( 3 mg/kg),Yiqi Huoxue Recipe group( 14.4,43.2,28.8 g/kg),each group of numble is eight.After the success of the model group,the western medicine group was treated with diltiazem hydrochloride,Chinese medicine group was treated with different doses of Yiqi Huoxue Prescription.45 days later,PCR was used to detect the changes of AC and PKA gene content in myocardium tissue under anesthesia.Results Compared with the model group,the AC and PKA concentrations were significantly decreased( P < 0.01); Compared with the western medicine group,the AC and PKA levels in the high-dose Chinese medicine group decreased significantly( P < 0.01).Conclusion The correlation between heart-Qi deficiency type of coronary heart disease and the changes of expression of AC and PKA gene content.The change of AC and PKA gene content reveals the microscopic mechanism of heart-Qi deficiency type of coronary heart disease
