苯和甲苯混合暴露对石化工人血清免疫指标及miRNA-146a,-155表达的影响

【摘要】 目的:探讨石化工人短时间混合暴露低浓度苯系物对免疫效应及相关miRNA表达的影响。方法:招募40名石化工人,收集其班前班后血样检测血清IgA、Ig G、IgM,IFN-γ、IL-1β、IL-2、IL-8、TNF-α及外周血单个核细胞miRNA-146a,-155,并检测工人作业期间个体苯系物水平。结果:根据外暴露检测结果将工人分为苯暴露组(20例)和对照组(20例)。苯暴露组甲苯检出水平显著高于对照组(P<0.05)。苯暴露组Ig G显著降低(P<0.05);苯和甲苯对IL-1β与TNF-α的下降有交互作用(P<0.05),但不影响miRNA的表达;miRNA-146a和miRNA-155对TNF-α的降低有交互作用(P<0.05)。结论:低浓度苯和甲苯短期混合暴露可降低机体免疫功能且可能受到细胞因子相关miRNA的调控。

【Abstract】 Objective To investigate the effects of immune system and immuno-related mi RNAs expressionin low-dose benzene and its homologue mixed-exposed workers in a short time. Methods A total of 40 workerswere recruited from a petrochemical factory,and their blood samples were collected before and after work todetect the levels of Ig A,Ig G,Ig M,IFN-γ,IL-1β,IL-2,IL-8,TNF-α in serum and mi RNA-146 a,-155 expressionin the peripheral blood mononuclear cell. In the meanwhile,we measured the individual benzene and its homologueexposure level of recruited workers. Results According to the concentration of benzene,the subjects were dividedinto benzene-exposed group(20 workers)and control group(20 workers). And toluene exposure level in benzene-exposed group was significantly higher than that in control group(P < 0.05). Significant decrease of Ig G was foundin benzene-exposed workers after one work shift,compared with control group(P < 0.05). Moreover,the interactionbetween benzene and toluene was significant correlated with the decrease of IL-1 β and TNF-α(P < 0.05).Additionally,the interaction between mi RNA-155 and mi RNA-146 a was significant correlated with the decrease ofTNF-α(P<0.05). Conclusions:Occupational exposure to low-dose benzene and toluene in a short time coulddecrease immune function. And there may be an effect of mi RNAs in the regulation of cytokine.
