注射用黄芪多糖体外诱导人鼻咽癌CNE-2细胞自噬及其机制
Effect of astragalus polysaccharides on autophagy in nasopharyngeal carcinoma cell line CNE-2
【摘要】 目的:探讨注射用黄芪多糖诱导人鼻咽癌细胞株CNE-2的自噬作用。方法:采用CCK8检测注射用黄芪多糖对CNE-2增殖的抑制作用;吖啶橙染色观察注射用黄芪多糖作用于CNE-2细胞引起的自噬形态变化;透射电镜观察注射用黄芪多糖作用于CNE-2细胞后引起的细胞超微结构变化。Western blot实验检测注射用黄芪多糖作用于CNE-2细胞后Beclin-1的表达变化。结果:注射用黄芪多糖对CNE-2细胞的生长有明显抑制作用,且具有浓度及时间依赖性。注射用黄芪多糖作用后可诱导CNE-2细胞发生自噬。透射电镜下细胞体内大空泡及膜性双层结构增多;吖啶橙染色可见注射用黄芪多糖作用后观察到更多的亮红色酸性自噬泡。Western blot结果表明Beclin-1蛋白表达增高。结论:注射用黄芪多糖抑制细胞增殖,可能通过上调beclin1表达诱导人鼻咽癌细胞CNE-2细胞发生自噬。
【Abstract】 Objective To investigate the effect of astragalus polysaccharides injection(APS) on autophagyof human nasopharyngeal cancer CNE-2 cells. Methods The inhibitory effect of APS on proliferation of CNE-2cells was measured by CCK8 assay. Morphological changes of autophagy of APS was detected by acridine orange(AO) staining. Transmission electron microscopy was perform to observe the morphological alterations in theautophagic cells. The expression of Beclin-1 protein was detected by Western blot assay. Results APS markedlyinhibited cell growth in a dose-and time-dependent manner. Increases of the number of large vacuoles and doublelayered membrane structure were observed by transmission electron microscopy. Results of AO staining revealedmore bright red cytoplasm or nucleus in the ceradime group,which proved the existence of acidic vesicularorganelles. Western blotting showed that Beclin-1 protein level was up-regulated. Conclusion APS may induceautophagy of human nasopharyngeal cancer CNE-2 cells via up-regulating the expression of apoptosis-related geneof beclin1 and inhibit the proliferation of CNE-2 cells.
