聚丙烯酰胺凝胶电泳法鉴别哈蟆油药材及其伪品

【摘要】 目的建立哈蟆油与伪品蟾蜍油、牛蛙油和青蛙油水溶性蛋白的非变性聚丙烯酰胺凝胶电泳(native-polyacrylamide gel electrophoresis,Native-PAGE)和十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(sodium dodecyl sulfate-polyacrylamide gel electrophoresis,SDS-PAGE)鉴别方法,并对2种电泳方法进行比较。方法采用Native-PAGE和SDS-PAGE两种电泳系统,分别对不同产地哈蟆油与其伪品的水溶性蛋白的组成及其分子量分布范围进行比较研究。结果 Native-PAGE实验结果显示:哈蟆油与桓仁林蛙油均具有6条特征谱带,黑龙江林蛙油具有5条特征谱带,而蟾蜍油、牛蛙油和青蛙油的特征谱带数目分别为2条、1条和1条。SDS-PAGE实验结果显示:哈蟆油、黑龙江林蛙油和桓仁林蛙油均具有11条特征谱带,而蟾蜍油、牛蛙油和青蛙油的特征谱带数目分别为5条、6条和6条。2种电泳图谱中,哈蟆油等正品药材与伪品蟾蜍油、牛蛙油和青蛙油在特征谱带位置等特征上有明显区别。结论 Native-PAGE和SDS-PAGE两种电泳方法均可用于哈蟆油与类似品和伪品的鉴别,为哈蟆油质量评价和控制方法提供了一种简易的鉴别方法。

【Abstract】 Objective To establish electrophoretic methods,including Native-PAGE and SDS-PAGE,to characterize the soluble proteins of Oviductus Ranae,its analogous and adulterant materials and make a comparison of both methods.Methods The Native-PAGE and SDS-PAGE methods were applied to characterize the composition of the protein and the range of molecule weight for soluble proteins of oviduct samples from five Rana sp.and Bufo bufo gargarizans collected from different habitats.Results The results showed that,in Native-PAGE patterns,6 protein bands with characteristic electrophoretic mobility were diagnostic to R.temporaria chensinensis,f ive bands differently to R.huanrenensis and R.amurensis,2 bands for Bufo bufo gargarizans,one band differently to R.catesbeiana and R.nigromaculata,respectively.In SDSPAGE patterns,11 similar polypeptides from R.temporaria chensinensis,R.amurensis and R.huanrenensis were characterized,while 5 polypeptides differently from B.bufo gargarizans and R.nigromaculata,and 6 from R.catesbeiana were diagnostic.The results show that there are significant difference in strength and position of soluble proteins between Ranae Oviductus,and its adulterant materials.Conclusions The electrophoretic systems of Native-PAGE and SDS-PAGE could be used to characterize Ranae Oviductus and its analogous and adulterant materials,which may provide a simple identification method for the quality control of Ranae Oviductus.
