低甾醇沙棘籽油制备工艺的研究

【摘要】 目的采用气相色谱法测定沙棘籽油中甾醇含量,考察低甾醇沙棘籽油(sea buckthorn seed oil,SBSO)的制备工艺。方法采用皂化萃取法、络合法、脱胶碱炼法和吸附法分别制备低甾醇沙棘籽油,以注射用大豆油中甾醇含量为对照,通过气相色谱法对沙棘籽油中甾醇含量进行检测。结果硅胶吸附法制备的低甾醇沙棘籽油总甾醇含量显著降低,与沙棘籽毛油相比,占总甾醇90%左右的β-谷甾醇由19.56%降至6.31%。皂化萃取法、络合法、脱胶碱炼制备的低甾醇沙棘籽油甾醇含量均有不同程度的降低,β-谷甾醇含量分别为13.46%、8.22%和9.36%。结论硅胶吸附法精制的沙棘籽油总甾醇含量显著降低,该制备工艺操作相对简易,除杂效率高。

【Abstract】 Objective To prepare low phytosterol sea buckthorn seed oil( LPSBSO) to make preparation for sea buckthorn seed oil-based parenteral fat emulsion( SSFE),and reference to the European Pharmacopoeia,determinate sterol content of SBSO using gas chromatography. Methods The LPSBSO were prepared by saponification extraction,complexometry, twice refining and silica gel adsorption method to decrease sterol content of SBSO,meanwhile determinate sterol content of SBSO using gas chromatography,and soybean oil for injection’ s sterol content as reference substance. Results LPSBSO for silica gel adsorption method prepared total sterol content was decreased significantly compared with the crude oil,especially, the content of β-sitosterol that accounts for at least 90 percent of total sterol content decreased from 19. 56% to 6. 31%compared with the crude oil. The β-sitosterol content of the LPSBSO prepared by aponification extraction,complexometry, twice refining and silica gel adsorption method was decreased in different degree to 13. 46%,8. 22% and 9. 36% respectively. Showed silica gel adsorption method was the relatively ideal preparation method. Conclusions The total sterol content of LPSBSO prepared silica gel adsorption method with operation easily and efficiently is decreased significantly.
