甲状腺乳头状癌发病风险和Bcl-2基因多态性的关系研究

【摘要】 目的探讨甲状腺乳头状癌(PTC)发病风险与B细胞淋巴瘤/白血病基因-2(Bcl-2)基因多态性的关系。方法选取2014年5月至2016年5月收治的65例PTC患者为观察组,同期选取72体检健康者为对照组。采集所有受试者外周血并提取DNA,利用限制性内切酶片段长度多态性PCR(PCR-RFLP)技术检测分析Bcl-2基因多态性,分析Bcl-2基因-938C/A多态性位点与PTC发病风险之间的关系。结果观察组与对照组Bcl-2基因-938C/A多态性位点基因型分布均符合Hardy-Weinberg平衡;观察组Bcl-2基因-938C/A多态性位点A/A基因型频率明显低于对照组,差异有统计学意义(P<0.05);观察组Bcl-2基因-938C/A多态性位点A等位基因频率显著低于对照组(P<0.05);观察组Bcl-2基因-938C/A多态性位点各基因分型频率与患者年龄、性别、肿瘤大小、包膜浸润、淋巴转移等无相关性(P>0.05),TNM不同分期中A/A基因型频率有显著差异,其中Ⅰ~Ⅱ期患者A/A基因型频率显著高于Ⅲ~Ⅳ期(P<0.05)。结论 Bcl-2基因-938C/A多态性位点与PTC发病有关,该基因位点A/A基因型以及A等位基因与PTC发病风险降低有关。

【Abstract】 Objective To investigate the relationship between the risk of Papillary thyroid carcinoma and the Bcl-2 gene polymorphism.Methods 65 PTC patients in our hospital were selected as the observation group,and 72 healthy patients were selected as control group. DNA was extracted from peripheral blood of all subjects,Bcl-2 gene polymorphism was detected by PCR-RFLP. The relationship between Bcl-2 gene-938 C/A polymorphism and the risk of thyroid papillary carcinoma were analyzed. Results Bcl-2 gene-938 C/A genotype distribution of the observation group and the control group were in accordance with Hardy-Weinberg balance; Bcl-2-938 C/A A/A genotype of the observation group was significantly lower than the control group,the difference was statistically significant( P < 0. 05). The A allele frequency was significantly lower than the control group( P < 0. 05). There was no correlation between the Bcl-2-938 C/A locus genotypes and the observation group of patients with age,gender,tumor size,capsule invasion,lymph node metastasis( P > 0. 05). A/A genotype in different TNM stages had significant differences,A/A genotype frequency of Ⅰ ~ Ⅱ patients was significantly higher than that in stage Ⅲ ~ Ⅳ( P < 0. 05). Conclusion Bcl-2-938 C/A polymorphism is related to the occurrence of PTC disease,and the A/A genotype and A allele are associated with a decreased risk of PTC.
