瑞舒伐他汀、普罗布考对Apo-E-/-小鼠心脏内LOX-1表达的影响

【摘要】 目的探讨瑞舒伐他汀、普罗布考及二者联合对载脂蛋白E基因敲除(Apo-E-/-)小鼠心脏内血凝集素样氧化低密度脂蛋白-1(LOX-1)表达的影响。方法雄性8周龄Apo-E-/-小鼠40只被随机分为对照组(单纯高脂饲料)、瑞舒伐他汀组(单纯高脂饲料+10 mg/kg灌胃)、普罗布考组(100 g饲料加入1 g普罗布考药粉)、联合组(10 mg/kg瑞舒伐他汀灌胃+100 g饲料加入1 g普罗布考药粉)4组,均采用相应的方法进行喂养,12周后进行解剖,显微镜下观察各组小鼠的主动脉脂质沉淀及动脉粥样硬化斑块情况,来判断动脉粥样硬化模型是否建立成功,取出心脏。应用实时定量荧光PCR及ELISA方法检测各组小鼠心脏中LOX-1在mRNA及蛋白水平的表达。结果显微镜下观察各组主动脉可见治疗组脂质沉积及粥样硬化斑块量较对照组少,以联合组为最少;对照组、普罗布考组、瑞舒伐他汀组、联合组4组小鼠心脏中LOX-1在mRNA及蛋白水平的表达量分别为:12.43±11.716,4.39±2.844;10.82±9.151,2.29±0.575;4.479±0.238,2.880±0.35及3.553±0.719,2.634±0.297,普罗布考组、联合组LOX-1在mRNA及蛋白水平明显低于对照组,差异具有统计学意义(P<0.05);但瑞舒伐他汀组与对照组相比差异无统计学意义(P>0.05)。结论瑞舒伐他汀、普罗布考及二者联合可减少主动脉中脂质沉积,但短期内对心脏中LOX-1的表达无明显影响。

【Abstract】 Objective To investigate the effects of rosuvastatin,probucol and their combination on the expression of LOX-1 in the heart of Apo-E-/-mice,and to explore the effects of two drugs on the expression of LOX-1 in the heart of the atherosclerosis model influences.Methods Forty male Sprague-Dawley rats were randomly divided into control group,rosuvastatin group,probucol group and combination group.The same method was used for feeding. After 12 weeks,under the microscope to observe aortic lipid deposition and atherosclerotic plaque,to determine whether the success of atherosclerosis model,and remove the heart. Real-time PCR and ELISA were used to detect the expression of LOX-1 mRNA and protein in the hearts of mice. Results The number of lipid deposition and atherosclerotic plaque in the treatment group were lower than those in the control group,the control group,probucol group,rosuvastatin group,combined group 4. The expression levels of LOX-1 in the heart of mice were: 12. 43 ± 11. 716,4. 39 ± 2. 844,10. 82 ± 9. 151,2. 29 ± 0. 575 and 4. 479 ± 0. 238,2. 880 ± 0. 351,3. 553 ± 0. 719 and 2.634 ± 0. 297 respectively. The mRNA and protein levels of LOX-1 in the combination group were significantly lower than those in the control group. The differences were statistically significant( P < 0. 05). But there was no significant difference between rosuvastatin group and control group( P > 0. 05). Conclusion Rosuvastatin,probucol and combination of the two can reduce lipid deposition in the aorta. There is no significant effect on the expression of LOX-1 in the heart in short term.
