【摘要】 目的探讨与细胞周期调控密切相关的纺锤体检查点基因MAD1和BUB3在自然流产组织中的表达情况及自然流产的发生机制。方法收集2015年5月至2016年5月稽留流产的标本,采用CNVplex高通量多重基因拷贝数检测技术将流产标本分为两组:50例有染色体异常的流产标本组(异常组),50例未见染色体异常的流产标本组(正常组)。采用RT-PCR检测两组胚胎绒毛组织中MAD1、BUB3的mRNA的表达情况。结果在稽留流产的胚胎中,与正常组相比,有染色非整倍体异常组MAD1的mRNA表达水平呈上调趋势,BUB3的mRNA表达水平呈下调趋势(P<0.05)。结论 MAD1及BUB3的正常表达对于胚胎发育十分重要,在自然流产染色体异常胚胎中MAD1及BUB3基因表达的改变,可能抑制SAC的激活,使得下游重要蛋白表达量异常,导致细胞分裂异常,产生胚胎染色体异常,继而诱发流产。

【Abstract】 Objective To explore the expression of closer related with cell cycle regulation of the spindle checkpoint gene( MAD1 and BUB3) in spontaneous abortion embryos and the mechanism of Unexplained Spontaneous Abortion embryos. Methods The samples of missed abortion between May 2015 and May 2016 of Huadu District People’s Hospital of Guangzhou were collected. The abortion samples were divided into two groups by using CNV plex high throughput multiplex gene copy number detection technique. A group included 50 cases of chromosome abnormalities. Another group included 50 cases of no chromosomal abnormalities. The expression of MAD1 and BUB3 mRNA in two groups of fetal chorionic tissues was detected by RT- PCR. Results Compared with nomal group,the mRNA expression level of MAD1 in chromosome abnormalities group was upward trend,the mRNA expression level of BUB3 was down regulated( P < 0. 05). Conclusion The normal expression of MAD1 and BUB3 is very important for embryonic development. Changes in expression of MAD1 and BUB3 gene in abnormal embryos of spontaneous abortion can inhibit the activation of SAC,Lead to abnormal expression of important downstream protein,and then cause abnormal cell division,generate embryonic chromosomal abnormalities,lead to abortion.
