益气活血养阴解毒方对大鼠放射性肺损伤的治疗作用
Efficacy of Yiqihuoxue-yangyinjiedu Recipe for Radiation-induced Lung Injury of Rats
【摘要】 目的探讨益气活血养阴解毒方对放射性肺损伤的治疗作用以及对血清转化生长因子-β1(TGF-β1)、肿瘤坏死因子-α(TNF-α)含量的影响。方法采用随机数字法,将90只大鼠分为A组(空白对照)、B组(照射)、C组(激素+照射)、D组(中药低剂量+激素+照射组)、E组(中药正常剂量+激素+照射组)、F组(中药高剂量+激素+照射组)六组,B~F组大鼠双肺均采用6MV X线照射,建立放射性肺炎模型。分别于放射双肺后的第7 d、14 d、21 d、28 d、60d随机断颈处死5只大鼠,获得右中肺进行肺部病理学评分,并采血采用ELISA法检测各时期大鼠血清TNF-α和TGF-β1含量。结果 B组各时期肺部损伤评分、血清TGF-β1和TNF-α含量均显著高于其他五组,E组和F组各时期肺部损伤评分、血清TGF-β1和TNF-α含量均显著低于C组;随着中药给药剂量的上升,肺部损伤评分、血清TGF-β1和TNF-α含量呈现下降趋势。结论与激素相比,加用益气活血养阴解毒方可有效降低大鼠放射性肺损伤、血清TGF-β1和TNF-α含量。
【Abstract】 Objective To investigate the efficacy of yiqihuoxue-yangyinjiedu recipe for radiation-induced lung injury of rats,and influence on transforming growth factor-β1( TGF-β1) and tumor necrosis factor-α( TNF-α) in serum lever of rats with radiation-induced lung injury. Methods 90 rats were randomly divided into the control group( group A),radiation group( group B),dexamethasone and radiation group( group C),low dose Chinese medicine and dexamethasone and radiation group( group D),normal dose Chinese medicine and dexamethasone and radiation group( group E),high dose Chinese medicine and dexamethasone and radiation group( group F). To establish radiation lung injury model,The rats from group B to group F were irradiated at whole lung with a dose of 18 Gy( 3 Gy/F,2 F/W). Execute 5 rats of every group at day7,day14,day21,day28,day 60 after radiation. Histopathologic score of right middle lobe were be done. The level of TGF-β1 and TNF-α in serum were detected by ELISA. Results The lung injury score,level of TGF-β1 and TNF-α at all time point of group B were significantly higher than the other groups’.Group E and F were significantly lower than the group C. With the rise of Chinese medicine dose,the lung injury score,level of TGF-β1 and TNF-α showed a downward trend. Conclusion Yiqihuoxue-yangyinjiedu recipe can effectively reduce the lung injury score,level of TGF-β1 and TNF-α of rats with radiation-induced lung injury,compared with the dexamethasone therapy group.
