吉西他滨对人非小细胞肺癌A549、H1299细胞增殖及凋亡的影响
The effects of Gemcitabine in human non-small cell lung cancer A549 and H1299 cells proliferation and apoptosis
【摘要】 目的:研究吉西他滨对人非小细胞肺癌A549及H1299细胞增殖及凋亡的影响。方法:应用MTT法及流式细胞技术测定不同浓度的吉西他滨对A549及H1299细胞体外增殖的抑制作用。结果:(1)MTT法检测不同浓度(1.25、2.5、5、10、20、40、60μg/ml)吉西他滨分别作用于A549及H1299细胞24、48、72h后,其对A549和H1299细胞的抑制作用逐渐增强,且具有一定的剂量和时间依赖性,但是随着时间的延长,吉西他滨对A549具有药物耐受性,而对H1299具有药物敏感性;(2)A549及H1299细胞随着培养时间的延长,细胞生长逐渐进入对数期,后进入平台期;(3)倒置显微镜下观察发现,A549及H1299对照组细胞贴壁生长,生长状态良好,形态大多数为多角形、或梭形,贴壁牢固,细胞间连接紧密。随着药物作用时间的增加,细胞逐渐皱缩、坏死、碎裂;(4)流式细胞仪检测不同浓度的吉西他滨(0、2.5、5、10、20、40μg/ml)对A549及H1299细胞凋亡具有明显的剂量依赖关系。结论:吉西他滨对A549及H1299细胞增殖有较强的抑制作用;呈现一定的浓度抑制作用和时间抑制作用。随着时间的延长,吉西他滨对A549细胞耐药,而对H1299细胞敏感。
【Abstract】 Objective:Study of the effects of Gemcitabine in human non-small cell lung cancer A549 and H1299 cells proliferation and apoptosis.Methods:Determined by MTT method and flow cytometry technology detect that various concentrations of Gemcitabine in A549 and H1299 cells proliferation in vitro.Results:(1)Determined by MTT method which detect different concentrations(1.25,2.5,5,10,20,40,60μg/ml)of Gemcitabine in A549 and H1299 cells after 24、48、72 hours.The result shows that the inhibition of A549 and H1299 cells gradually strengthened,and which has a certain dose and time dependent characteristic,but when the incubation time was prolonged,which indicates that Gemcitabine was resistant to the A549,which was sensitive to H1299 cells.(2)When the incubation time was prolonged,A549 and H1299 cells gradually into the logarithmic growth period,after go in plateau.(3)Observed under inverted microscope,the control group of A549 and H1299 which grow against the wall in good state.The mostly form which were diamond or spindle,those cells tightly attached and tight junctions between cells with the increase of drug effect time,cells gradually shrinking、necrosis and rupture.(4)How cytometry instrument detect that various concentrations of gemcitabine(0,2.5,5,10,20,40μg/ml)in A549 and H1299 cells apoptosis has obvious dose response relationship.Conclusion:Gemcitabine has quite strong inhibitory effect on cells proliferation,which presents certain concentration dose and time dependent.With the time goes by,gemcitabine is resistant to the A549 and sensitive to H1299 cells.
