人重组组织因子微颗粒的制备研究
Study the Preparation of Recombinant Human Tissue Factor Microparticles
【摘要】 采用磷脂酰胆碱(DOPC)、磷脂酰丝氨酸(DOPS)2种合成磷脂,按照6∶4、7∶3、8∶2的比例(摩尔比),分别以磷脂质量浓度为2.0%、2.5%、3.0%的混合溶液,超滤交换,制备9种双层磷脂微颗粒(MP),MP与人重组组织因子(rhTF243)脂化,得到人重组组织因子的磷脂微颗粒(rhTF243+MP)。经PT方法检测比较9种rhTF243+MP的凝血活性、稳定性及储存时间,确定最佳磷脂配比、浓度及制备工艺,制备具有稳定促凝血活性的rhTF243+MP。
【Abstract】 The two kinds of synthetic phospholipids,Phosphatidylcholine(DOPC)and phosphatidylserine(DOPS),were mixed according to the ratio of 6∶4,7∶3,8∶2(molar ratio) and 2%,2.5%,3% mass concentration,exchanged by ultrafiltration method,maked 9 kinds of microparticles(MP). MP and recombinant human tissue factor(rhTF243)were combined and getten recombinant human tissue factor(rhTF243+ MP). The clotting activity,stability and storage time of 9kinds of TF243+MP were was completed. The rhTF243+MP can provide the raw materials for the production of PT reagent products,provide the standardization for the standardization of PT reagents,and provide the assistance for the study of coagulation mechanisms and disease treatment.
