ApoE与β淀粉样蛋白在阿尔茨海默病中相互作用的研究进展
The Recent Advance in Interactions between Apolipoprotein E and Amyloid-beta in Alzheimer’s Disease
【摘要】 阿尔茨海默病(Alzheimer’s disease,AD)是一种中枢神经系统神经退行性疾病,至今尚未明确其发病机制,但基于其典型的病理特征之一是β淀粉样蛋白(amyloid-beta,Aβ)聚集,Aβ沉积假说一直都是研究的重点。近年来,载脂蛋白E(apolipoprotein E,APOE)对Aβ代谢清除的影响备受关注。研究表明,APOEε4等位基因是散发性AD的危险因素,且APOE对Aβ具有很高的亲和力,不同亚型的APOE对Aβ的代谢有不同的影响,这为对AD的认识、防止及治疗提供了新的研究方向。
【Abstract】 Alzheimer’s disease(AD) is a neurodegenerative disease of the central nervous system. Its pathogenesis is still incompletely understood. The accumulation of amyloid-beta(Aβ) is one of the typical pathologic symptoms, and the "amyloid hypothesis" has dominated Alzheimer research for a long time. Recently, the effect of apolipoprotein E(APOE) in Aβ metabolism attracts extensive concern. It suggests that ε4 allele is a risk factor in sporadic Alzheimer disease. Apolipoprotein E has a high affinity with Aβ, while different isoforms of apolipoprotein E have different influences on Aβ metabolism. These results provide a new way for the identification, prevention and therapy of AD.
