补骨脂素对中波紫外线导致人皮肤HaCaT细胞光老化的保护作用
Protective Role of Psoralen Grain of on Light Aging of Human Skin HaCaT Induced by UVB
【摘要】 目的:研究补骨脂素对中波紫外线(UVB)导致人皮肤HaCaT细胞光老化的保护作用及其作用机制。方法:选择不同浓度的补骨脂素,MTT法筛选药物的浓度;使用中波紫外线(UVB)照射永生化的HaCaT细胞建立UVB光老化模型;使用三种不同浓度的补骨脂素处理光老化模型,MTT法检测细胞的增殖及氧化试剂盒检测细胞中氧化酶的活性。RT-PCR及Western Blot分别检测JNK和白介素-8(IL-8)mRNA及蛋白表达量。结果:与空白组相比,10-7mol/L、10-6mol/L、10-5mol/L补骨脂素组对HaCaT具有无明显的增殖作用(P>0.05);与模型组相比,10-7mol/L、10-6mol/L、10-5mol/L补骨脂素组对HaCaT具有无明显的增殖作用(P>0.05),但是10-7mol/L、10-6mol/L、10-5mol/L补骨脂素组SOD、GSH、CAT活性升高(P<0.01),细胞JNK、IL-8 mRNA表达量均降低(P<0.01),细胞JNK、IL-8蛋白表达量均降低(P<0.05或P<0.01)。结论:补骨脂素能够显著的保护HaCaT细胞的光老化,其机制可能与增强抗氧化酶活性,及抑制JNK信号通路,减少炎症因子的分泌有关。
【Abstract】 Objective: To study the protective role of psoralen grain on light aging of human skin HaCaT cells induced by UVB rays(UVB) and its underlying mechanism. Methods: The different concentrations of psoralen were selected and the suitable concentration of the drugs was screened by MTT method. Using UVB rays(UVB) radiation, immortalized HaCaT cells UVB light aging model was established, and then treated with three different concentrations of psoralen processing. The cell activity was determined by MTT method and oxidase activity in the cell was detected by kits, and the expression of JNK and interleukin 8(IL-8) mRNA and protein was respectively detected by RT-PCR and Western Blot. Results: Compared with blank group, 10-7mol/L, 10-6mol/L, 10-5mol/L psoralen grain hadn,t obvious effect on the proliferation of HaCaT cells(P>0.05); compared with UVB model group, the cell proliferation in psoralen group had no significant change(P>0.05), but after treatment with 10-7mol/L, 10-6mol/L, 10-5mol/L psoralen, the activity of SOD and GSH and CAT was increased(P<0.05), and the expression of JNK and IL-8 mRNA was decreased(P<0.01), and the expression of JNK and IL-8 protein was also decreased(P<0.05 or P<0.01). Conclusions: The psoralen grain can significantly protect HaCaT cells light aging, and its mechanism may be related with increasing the antioxidase activity, inhibiting JNK signaling pathways, and reducing the secretion of inflammatory cytokines.
