壳寡糖-赖氨酸美拉德反应产物的优化制备及抗氧化活性
Optimized Preparation and Antioxidant Activity of Maillard Reaction Products Derived from Chitooligosaccharide and Lysine
【摘要】 以壳寡糖(COS)和赖氨酸(Lys)为反应物,通过单因素试验和响应面法优化制备壳寡糖-赖氨酸美拉德反应产物(CL-MRPs),测定其抗氧化活性并进行荧光和红外表征。结果表明:最优制备条件为反应时间118 min,反应温度114℃,pH 4.5,COS与Lys摩尔比2.3:1。此条件下制备的CL-MRPs的铁离子还原/抗氧化力为13.86,对ABTS自由基、超氧自由基、DPPH自由基和羟基自由基的清除率分别达到79.97%,84.93%,94.17%和97.48%,表现出强抗氧化活性。CL-MRPs的荧光强度明显高于反应物,说明有荧光产物形成。CL-MRPs的红外光谱表明反应过程中有席夫碱化合物形成。
【Abstract】 The preparation of chitooligosaccharide-lysine Maillard reaction products(CL-MRPs) was optimized by one-factorat-a-time experiment and response surface methodology using chitooligosaccharide(COS) and lysine(Lys) as reactants.The antioxidant activity of CL-MRPs was tested,and CL-MRPs were characterized by fluorescence and FTIR spectra.The results showed that the optimum reaction conditions were as follows:reaction time of 118 min,reaction temperature of 114 ℃,pH 4.5 and mole ratio of COS and Lys of 2.3:1.Under the optimum conditions,the ferric reducing/antioxidant power of CL-MRPs was 13.86;the scavenging activity of CL-MRPs reached 79.97%,84.93%,94.17% and 97.48% for ABTS,superoxide,DPPH and hydroxyl radicals,respectively.These results suggested that CL-MRPs had strong antioxidant activity.The fluorescence intensity of CL-MRPs was higher than that of reactants,indicating the production of fluorescence compounds.The FTIR spectra of CL-MRPs showed the formation of Schiff base compounds.
