蔬菜中色素检测及在卷烟中的应用

【摘要】 建立了玉米及胡萝卜中叶黄素、玉米黄素、β-隐黄素、α-胡萝卜素和β-胡萝卜素反相高效液相色谱方法。玉米及胡萝卜经皂化和提取后以Waters YMCTM Carotenoid 3μm色谱柱(4.6 mm×150 mm)分离,450 nm波长DAD检测;流动相A相为乙腈;流动相B相为乙酸乙酯,梯度洗脱,流速为1.0 mL/min,柱温30℃。在此检测条件下,得到不同提取工艺的玉米及胡萝卜色素的检测结果,并对检测方法进行考察;结果表明,玉米及胡萝卜中5种色素总含量以亚临界萃取最高,亚临界萃取法提取玉米及胡萝卜色素的工艺最为适宜。卷烟加香结果表明,玉米和胡萝卜色素可明显提高香气质、掩盖杂气、改善烟气细腻柔和程度、增加烟气回甜等。

【Abstract】 HPLC method of detecting lutein, zeaxanthin, β-cryptoxanthin, α-carotene and β-carotene in corn and carrot was established. After saponification and extraction, corn and carrot were separated by Waters YMCTM Carotenoid 3 μm column(4.6 mm×150 mm) and detected by DAD with wavelength of 450 nm. Mobile phase A was acetonitrile and mobile phase B was ethyl acetate, and gradient elution with the flow rate of 1.0 mL/min, with column temperature of 30 ℃. On such condition, 5 kinds of pigments including lutein(Lut), zeaxanthin(Zea), β-cryptoxanthin(β-Cryp), α-carotene(α-C) and β-carotene(β-C) in corn and carrot extracted by different methods were determined, and the effectiveness of testing methods was investigated. The results showed that total content of 5 kinds of pigments was the highest when using subcritical extraction, which could keep the pigments in corn and carrot maximum. From the aspects of increasing pigment content and retaining material properties, it was the best to choose subcritical extraction to extract corn and carrot. Through their addition in cigarette, the aroma quality could be improved; Meanwhile, miscellaneous gas could be covered, soft and delicate degree of cigarette could be improved, and aftertaste of sweetness could be increased.
