花生中黄曲霉毒素B1提取方法的优化

【摘要】 主要研究了酶联免疫法检测花生中黄曲霉毒素B1的提取条件,通过单因素试验分别研究料液比、甲醇体积分数、提取时间和超声波功率对黄曲霉毒素B1提取效果的影响,通过正交试验法确定花生中黄曲霉毒素B1的最佳提取条件。结果表明,各因素影响花生中提取黄曲霉毒素B1含量的大小为:超声波功率>甲醇体积分数>提取的时间>物料比。最佳提取条件为:料液比为1︰5 g/m L,甲醇体积分数60%,提取时间为11 min,超声波功率70 W。试验以为花生中黄曲霉毒素B1的检测提供简便、可靠、准确的分析方法。

【Abstract】 ELISA on the detection of AFB1 in peanut was mainly researched. In the single factor analysis, the effects of solid-liquid ratio, methanol concentration, extraction time and wave power were measured. The orthogonal test was used to optimize the conditions of extraction. The results showed that the order of extraction factor was as followings: wave power>methanol concentration>extraction time>solid-liquid ratio. The optimum conditions were solid-liquid 1︰5 g/m L, methanol concentration 60%, extraction time 11 min, wave power 70 W. The study presented a simple reliable accurate method for the detection of AFB1 in peanut.
