【摘要】 mTOR信号通路已被确定为几种癌症的致癌因素。癌症生物学的一个主要领域是基因组生物标记的发展它可以测量pi3k-akt-mtor途径的活动水平。PI3K-Akt-mTOR信号通路包含许多信号蛋白,包括Ras,PTEN,PIP,PI3K,AKTand mTOR。作为部分信号通路功能的串联功能是是由生长因子结合在细胞膜上的受体酪氨酸激酶引起的。重要的是PI3K-Akt-mTOR信号通路在癌症细胞的生长,增殖,以及生存有重要的作用,在众多癌症中被过度表达包括乳腺癌,卵巢癌和胰腺癌等。因此开发PI3K抑制剂不仅可以消灭肿瘤的生长,也可以协同其它的标记物治疗和化疗治疗相关的癌症。由于这条信号通路重要,已经有了几种相关基因组的方法来评估细胞系中PI3K-Akt-mTOR信号通路的活性以及原发性患者肿瘤的活动情况。

【Abstract】 The mTOR signaling pathway has been identified as a key driver of carcinogenesis in several cancer types. A major area of focus in cancer biology is the development of genomic biomarkers that it can measure the activity level of the pi3k-akt-mtor pathway.The PI3K-Akt-mTOR pathway is composed of a number of proteins, including Ras, PTEN, PIP, PI3K, AKtand mTOR. The pathway functions as part of a signaling cascade initiated by the binding of growth factors to receptor tyrosine kinases are located on the cell membrane. Importantly, the PI3K-Akt-mTOR pathway plays a central role in cancer cell growth, proliferation, and survival, and is over expressed in numerous cancers including breast, ovarian, and pancreatic cancer. Consequently, there have been significant efforts in developing PI3K inhibitors that can abrogate tumor growth and act synergistically with other targeted therapy and chemotherapy.Due to the importance of this pathway, several genomics-based approaches have been introduced to assess the activity of the PI3K-Akt-mTOR pathway in cell lines and in primary patient tumors.
