肾细胞癌组织中RASSF10基因表达及其启动子区甲基化变化的意义

【摘要】 目的观察肾细胞癌(RCC)组织中RASSF10基因表达及其启动因子区甲基化的变化,并探讨其意义。方法采用RT-PCR技术、甲基化特异性聚合酶链反应检测52例RCC组织及其相应癌旁组织、正常组织中RASSF10 mRNA相对表达量及其甲基化率,并分析RASSF10基因表达变化、启动子区甲基化与RCC患者病理特征的关系,分析RASSF10基因启动子区甲基化与其表达的关系。结果 RCC组织中RASSF10 mRNA相对表达量、甲基化率与癌旁及正常组织比较,P均<0.05。RASSF10基因表达及其启动子区甲基化率与患者年龄、病理类型、肿瘤大小、Fuhrman分级无关(P>0.05),与肿瘤TNM分期有关(P均<0.05)。RASSF10基因启动子区甲基化的RCC患者RASSF10 mRNA相对表达量与未甲基化患者比较,P<0.05。结论 RCC组织中RASSF10基因低表达可能与其启动子区高甲基化有关;RASSF10基因低表达及其启动子区甲基化可能参与了RCC的发生发展。

【Abstract】 Objective To observe the expression of RASSF10 gene in renal cell carcinoma( RCC) and its promoter methylation,and to investigate its significance. Methods The mRNA expression and methylation rate of RASSF10 gene in RCC tissues,para-carcinoma tissues,and normal tissues of 52 cases were detected by using reverse transcription polymerase chain reaction( RT-PCR) and methylation-specific polymerase chain reaction( MSP). Their relationship with pathological features of RCC patients and the relationship between mRNA expression and methylation of RASSF10 gene were analyzed. Results The mRNA relative expression level of RASSF10 gene in RCC tissues was lower,and the methylation rate was higher than that in the para-carcinoma tissues and normal tissues( both P < 0. 05). The mRNA expression and promoter methylation rate of RASSF10 gene in RCC tissues were not correlated with age,histological type,tumor size,and Fuhrman grade( all P > 0. 05),but were correlated with tumor TNM grade( P < 0. 05). The mRNA relative expression of RASSF10 gene in RCC patients with methylation was lower than that without methylation( P < 0. 05). Conclusion The low expression of RASSF10 gene in RCC tissues may be related to promoter hypermethylation,and low expression of RASSF10 gene and its promoter methylation may participate in the occurrence and development of RCC.
