正交试验优化板栗中硒蛋白的提取工艺

【摘要】 本实验优选恩施市富硒板栗,采用磷酸盐缓冲液浸提法提取硒蛋白,正交试验法优化板栗硒蛋白的提取条件,采用考马斯亮蓝G-250试剂结合法测定蛋白含量,原子荧光法测定硒含量。结果表明,影响富硒板栗含硒蛋白提取率的主次因素为:p H值>料液比>提取时间>提取温度,富硒板栗中含硒蛋白提取的最佳工艺条件是:p H值8.2,提取温度50℃,提取时间1 h,料液比1∶55。富硒板栗干粉中的总硒含量为2.79μg/g,富硒板栗蛋白粉中的硒含量为5.125μg/g。本研究旨在为恩施富硒板栗的精深加工和综合利用奠定了理论和技术基础。

【Abstract】 Enshi Selenium-enriched chestnut was used as materials to extract selenoprotein by phosphate buffer extraction method and to optimize the extraction process of selenoproteins by using the orthogonal experiment. Proteins and selenium content was measured by coomassie brilliant blue G-250 reagent and AFS( atomic fluorescence spectrometry) respectively. Results showed that the factors affecting extraction rate was as follows: p H > ratio of solid-liquid > extraction time > extraction temperature. The optimum extraction conditions of selenoprotein from chestnuts were p H 8. 2,extraction temperature 50℃,extraction time 1 hour and the ratio of solid-liquid 1∶ 55. The selenium content in Selenium-rich chestnut powder and Selenium-rich chestnut protein powder was 2. 79 μg/g and 5. 125 μg/g respectively. The aim of this study was to lay a theoretical and technical foundation for the intensive processing and comprehensive utilization of Se-enriched chestnut.
