沙利度胺联合顺铂对人宫颈癌裸鼠增殖的影响及其作用机制
【摘要】 目的:探讨沙立度胺联合顺铂对人宫颈癌移植瘤的生长的影响,观察沙利度胺对宫颈癌Hela细胞VEGF的影响。方法:建立宫颈癌裸鼠荷瘤模型,分为4组,每组10只。(1)沙利度胺组:200mg·kg-1·d-1,0.2ml腹腔内注射;(2)顺铂组:2mg·kg-1·d-1,0.2ml腹腔内注射;(3)联合组:沙利度胺:200mg·kg-1·d-1,顺铂:2mg·kg-1·d-1,各0.2ml腹腔内注射;(4)对照组:生理盐水0.2ml腹腔注射。给药2周后,观察各组裸鼠体重、计算肿瘤抑制率,采用免疫组化法检测沙利度胺处理后移植瘤组织中血管内皮生长因子(VEGF)。结果:用药2周后,(1)沙利度胺组、联合用药组、顺铂组肿瘤抑制率分别为39.20%、59.06%、34.13%,联合用药组抑瘤作用最显著(P<0.05),(2)与对照组相比,沙利度胺组作用于人宫颈癌Hela细胞后,其VEGF的表达明显低于未加药组,差异显著(P<0.05)。结论:沙利度胺与顺铂联合,可增加宫颈癌Hela皮下移植瘤的抑瘤率,沙利度胺可降低肿瘤内的VEGF的表达,抑制宫颈癌血管形成。
【Abstract】 Objective: To investigate the effect of thalidomide combined with cisplatin on the growth of human cervical cancer xenografts, observe the effects of thalidomide on VEGF cervical cancer cell line Hela. Methods: The nude mice bearing tumor model of cervical cancer were divided into 4 groups, each group had 10 mice.(1) Thalidomide group: 200 mg·kg-1·d-1 by intraperitoneal injection of 0.2 ml;(2) Cisplatin group: 2 mg·kg-1·d-1 by intraperitoneal injection of 0.2 ml;(3) Joint group: 200 mg·kg-1·d-1 of thalidomide and 2 mg·kg-1·d-1 of cisplatin by intraperitoneal injection of 0.2 ml;(4) Control group: normal saline by intraperitoneal injection of 0.2 ml. Two weeks after the treatment, observation group nude mice body weight, tumor inhibition rate calculation, using immunohistochemical method to detect gilad amine treatment after transplanted tumor tissue of vascular endothelial growth factor(VEGF). Results: Two weeks later.(1) Thalidomide group, combination group, cisplatin group tumor inhibition rates were 39.20%, 59.06%, 34.13%, but the combination group had the most significant inhibitory effect(P<0.05).(2) Compared with the control group, the degree of thalidomide group to act on human cervical cancer Hela, the expression of VEGF was lower than that in group without medicine, significant difference(P<0.05). Conclusion: Thalidomide combined with cisplatin, which can increase the Hela cervical cancer subcutaneous transplantation tumor inhibition rate. The expression of thalidomide can reduce the intratumoral VEGF, and inhibiting vascular formation of cervical cancer.
