补肾活血汤含药血清对骨髓间充质干细胞迁移及CXCR4表达的影响

【摘要】 【目的】探讨补肾活血汤对骨髓间充质干细胞(BMSCs)迁移及趋化因子受体CXCR4表达的影响。【方法】以大鼠BMSCs为研究对象,采用全骨髓贴壁法培养BMSCs。采用流式细胞术鉴定细胞表面抗原CD90、CD34、CD11b;采用Transwell试验检测不同剂量组补肾活血汤含药血清对BMSCs迁移的影响;采用Western blot法检测细胞CXCR4的蛋白表达水平。【结果】细胞表面抗原标记结果显示所得细胞为高纯度BMSCs。Transwell结果显示,补肾活血汤含药血清可显著增强大鼠BMSCs的迁移能力;Western blot结果显示,高、中、低剂量组补肾活血汤含药血清呈浓度依赖性上调大鼠BMSCs的CXCR4蛋白表达,与空白血清组比较,差异均有统计学意义(P<0.05)。【结论】补肾活血汤含药血清可促进BMSCs的体外迁移,并影响CXCR4蛋白的表达,其机制可能与上调CXCR4的表达、激活SDF-1/CXCR4轴有关。

【Abstract】 Objective To explore the effect of Bushen Huoxue Decoction(BHD)on the migration of bone marrow mesenchymal stem cells(BMSCs),and to explore its mechanism. Methods BMSCs were used as the cell model,and were cultivated by bone marrow adherent method. The cell surface antigens of CD90, CD34, and CD11 b were indentified by flow cytometry. Transwell assay was used to observe the effects of serum containing different dosages of BHD on cell migration of BMSCs. Western blotting method was used to measure the expression level of CXCR4 protein. Results The identification results for cell surface antigens indicated that the obtained cells were pure BMSCs. Transwell assay results showed that the vitality of BMSCs migration after treatment with serum containing BHD was superior to that of the blank serum group. Western blot results showed that the protein expression level of CXCR4 was enhanced by serum containing high-, middle-, and low-dosage BHD in concentration-depending mode(P < 0.05 compared with the blank serum group). Conclusion The serum containing BHD can promote BMSCs migration, and regulate the expression level of CXCR4 protein, and its mechanism may be related with the up-regulation of CXCR4 expression and with the activation of the SDF-1/CXCR4 axis.
