HCMV感染对下调ATF5的人胶质瘤U251细胞干性的影响
The Effect of HCMV Infection on the Stemness of Down-regulated ATF5 U251 Cells
【摘要】 研究HCMV感染对下调ATF5的U251细胞干性标记物CD133的表达、细胞增殖能力和克隆形成能力的影响。采用荧光定量PCR及Western-blot方法检测HCMV(MOI=1)感染的U251细胞IE、CD133基因及蛋白表达水平的变化,结果表明,随IE表达量的增加U251细胞中CD133表达量显著增加;成功构建沉默ATF5细胞系siATF5-U251,HCMV感染siATF5-U251细胞后使细胞增殖活性减弱,克隆形成率降低;荧光定量PCR及Western-blot检测HCMV感染显著抑制了siATF5-U251细胞内CD133的表达。因此,抑制ATF5的表达可成为恶性胶质瘤预防和治疗的新的作用靶点。
【Abstract】 To study the effect of HCMV infection on the expression of CD133,cell proliferation and clone formation of U251 cells,which were down regulated the ATF5 expression.The changes of IE,CD133 gene and protein expression levels of HCMV(MOI=1)infected U251 cells were detected by fluorescence quantitative PCR and Western-blot methods.The results showed that the expression of CD133 increased significantly with the increase of IE expression in U251 cells.Silencing ATF5 cell line siATF5-U251 was successfully constructed,and HCMV infection reduced the cell proliferation activity,and decreased the colony formation rate.The results of fluorescence quantitative PCR and Western-blot showed that HCMV infection significantly inhibited the expression of CD133 in siATF5-U251 cells.Therefore,inhibiting the expression of ATF5 may be a new target for prevention and treatment of malignant glioma.
