新型补铁剂—蔗糖铁的合成

【摘要】 研究新型补铁剂——蔗糖铁(Ⅲ)复合物的合成工艺路线。以三氯化铁为原料,水解形成氢氧化铁胶泥,再与蔗糖络合,形成蔗糖铁复合物。通过单因素实验确定最佳工艺条件。通过置换碘量法测定铁含量,借助高效液相色谱测定了蔗糖铁的分子量及分子量分布。三氯化铁与碳酸钠的质量比1:0.62,加料时间为1h,反应温度15℃~20℃时,氢氧化铁胶泥的产率最高。铁与蔗糖质量比1:14.8,反应时间10h,反应温度110℃~115℃,6 M氢氧化钠溶液调节pH值11.3~11.7,制得的蔗糖铁原料药溶液的重均分子量(Mw)为34 000~60 000;分布系数(MW/Mn)小于1.7;通过阴、阳离子交换,产物中Cl~-的含量控制在0.012%~0.025%之间,可滴定碱指标合格;0.8μm的滤膜过滤,分散体系均匀。该制备工艺简单可行,制得的产品含铁量高、分散均匀。

【Abstract】 To study preparation process of a new kind of iron-supplement of iron sucrose complex.Ferric hydroxide was prepared by hydrolysis using ferric chloride as starting material.And iron sucrose complex was synthesized by the reaction of ferric hydroxide with sucrose.The preparation parameters of iron sucrose complex were optimized by means of the single factor design.The content of iron in complex was determined with iodometry.The molecular weight and molecular weight distribution of sucrose iron were determined by HPLC.Optimum conditions for the preparation of ferric hydroxide is as follows:mass ratio of ferric chloride to sodium carbonate 1:0.62,reaction time 1 h,temperature 15~20℃;the optimum conditions for synthesis of iron sucrose:mass ratio of ferric hydroxide to sucrose 1:14.8,temperature 110~115 ℃,reaction time 10 h,pH 11.3 ~ 11.7.The average molecular weight(Mw)of the iron sucrose solution ranged from 34000 to 60000,the distribution coefficient(Mw/Mn)is less than 1.7.The Cl~-content in the product was controlled at 0.012% ~0.025% and the index of titratable base is qualified.The dispersion system was uniform with by 0.8μm membrane filtering.The preparation process of iron sucrose complex was feasible.The product is in uniform dispersion and its iron content is high.
