V_γ4~+γδT细胞清除可缓解咪喹莫特诱导的小鼠银屑病样皮炎
Depletion of V_γ4~+γδ T cells alleviates imiquimod-induced psoriasis-like skin inflammation in mouse model
【摘要】 目的观察V_γ4~+γδT细胞在咪喹莫特乳膏诱导的Balb/c小鼠银屑病模型中的作用。方法雌性8周龄Balb/c小鼠随机分为野生型组和V_γ4清除型组。咪喹莫特乳膏诱导银屑病模型后,采用PASI评分标准分别评判2组小鼠银屑病模型皮损的严重程度,HE染色观察皮损组织形态学改变,流式细胞仪分别检测2组小鼠正常及炎症时真皮组织中V_γ4T细胞的比例,Western blot检测2组小鼠正常及炎症时真皮组织蛋白IL-17、IL-23、IL-1β的表达水平。结果连续6 d涂抹0.5%咪喹莫特乳膏后,野生型组小鼠背部脱毛区域出现红斑、鳞屑、斑块的典型银屑病样皮损比V_γ4清除型组小鼠严重。流式细胞仪检测到野生型组正常小鼠真皮细胞内在少量的V_γ4T细胞,炎症时该细胞数量增多,V_γ4清除型组正常及炎症小鼠均无V_γ4T细胞表达。同时Western blot检测到V_γ4清除组小鼠真皮组织蛋白IL-17、IL-23、IL-1β炎性因子的表达水平也低于野生型组。结论清除小鼠体内V_γ4T细胞后,咪喹莫特诱导的银屑病样皮炎减轻,同时IL-17、IL-23、IL-1β3种炎性因子的蛋白表达水平均降低。
【Abstract】 Psoriasis,a common chronic inflammatory skin disease,is closely related with the IL-17 production.Previous studies have confirmed that the IL-17-producing γδT cells play a central role in the development of psoriasis.This study was designed to investigate the function of V_γ4~+T cells that belong to subset ofγδT cells in the model of psoriasis induced by imiquimod(IMQ) cream.Eight weeks old female Balb/c mice were randomly divided into wild-type group and V_γ4T cells eliminated group which was obtained by the injection of V_γ4 depletion antibodies for three days.The psoriasis-like skin inflammation was induced by daubing IMQ cream on the dorsal skin once a day for 6 successive days.The severity of the psoriasis skin was evaluated by the PASI score and the skin tissue morphology change was observed by HE.The dermal V_γ4T cells were detected by FACS.In addition,the expression of IL-17,IL-23 and IL-1β in dermal tissue were measured by Western blot.As a result,the wild-type group had a set of symptoms in the hair removal area like erythema,scales,which the PASI score was higher than the V_γ4T cells eliminated group.Besides,the protein levels of IL-17,IL-23,IL-1β in the dermal tissue were significantly decreased in the V_γ4T cells eliminated group.Our results suggested that eliminating V_γ4 T cells alleviate the imiquimod-induced psoriasis-like skin inflammation.
