SSR标记检验玉米杂交种子纯度核心引物的构建

【摘要】 利用50对SSR引物对收集到的18个玉米杂交种及其亲本进行SSR标记分析,以期筛选一套适于玉米杂交种纯度鉴定的核心SSR引物。试验结果表明,50对SSR引物在18个杂交种及其亲本中,有20对引物扩增出多态性特异条带,进一步对20对引物进行重复试验和PCR反应条件的优化,筛选出了8对SSR标记引物,分别在18个玉米杂交种及其亲本中扩增到了条带清晰、多态性好的条带,可用于其玉米杂交种子纯度鉴定和检验。这为玉米杂交种种子真实性和纯度检验、解决玉米种子质量纠纷提供了更为准确可靠的参考依据。

【Abstract】 18 maize hybrids and their parents were analyzed by 50 pairs of SSR marker primers in order to screen a set of core SSR primers suitable for purity identification of maize hybrids. The experimental results suggest that there are 20 pairs of primers amplified polymorphic specific bands in 18 hybrids among 50 pairs of SSR primers. Further optimization experiments were repeated and PCR reaction conditions by 20 pairs of primers. There are 8 pairs of SSR primers respectively in 18 maize hybrids and amplified the clear and polymorphic bands. They can be used for the hybrid maize seed purity identification and inspection. This will be a more accurate and reliable reference for the authenticity and purity test of Corn Single Cross seeds and the resolution of corn seed quality disputes.
