白蜡及白蜡高级烷醇对人真皮乳头细胞生长的影响
Effect of White Wax and Policosanol from White Wax Tween Aqueous Solution on HFDPCS
【摘要】 [目的]明确白蜡及白蜡高级烷醇吐温水溶液对人真皮乳头细胞(HFDPCs)生长的影响。[方法]采用台盼蓝法测定细胞的的接种数量,接种第2天给药,给药48 h后采用MTS法研究不同浓度吐温80、白蜡及白蜡高级烷醇吐温水溶液对人真皮乳头细胞的作用。[结果]人真皮乳头细胞在吐温80用量1.56 6.25μg·m L-1范围内能正常生长。在安全范围内,白蜡(增溶量为0.31 1.25μg·m L-1)吐温水溶液、白蜡高级烷醇(增溶量为3.12 12.5μg·m L-1)吐温水溶液均能促进人真皮乳头细胞的增殖,其中,水溶液中含有6.25μg·m L-1吐温80和1.25μg·m L-1白蜡、水溶液中含有6.25μg·m L-1吐温80和12.5μg·m L-1白蜡高级烷醇时细胞的增殖率明显高于非那雄胺。[结论]吐温80作为增溶剂,其用量低于6.25μg·m L-1时对人真皮乳头细胞安全。在保证吐温80对细胞安全的条件下,白蜡及白蜡高级烷醇可显著促进人真皮乳头细胞的增殖。
【Abstract】 [Objective]To investigate the cell proliferation effect of white wax and policosanol from white wax tween aqueous solution in human dermal papilla cells( HFDPCs). [Method]The cell seeding density was decided by Trypan blue test. Next day,change fresh medium which including treatment agent for HFDPCs. MTS assay was used to compare the proliferation effects of different concentrations of tween80,white wax and policosanol from white wax tween80 aqueous solutions on HFDPCs tween aqueous solutionafter 48 h. [Result]HFDPCs could grow wellwhenthe concentration of tween 80 was 1. 56- 6. 25 μg·m L-1. In the concentration range,white wax( solubility in water was from 0. 31-1. 25 μg·m L-1) and policosanol from white wax( solubility in water was 3. 125-12. 5μg·m L-1) tween aqueous solutions had the cell proliferation effect for HFDPCs. In which,the white wax aqueous solution including 6. 25 μg·m L-1tween80 and 1. 25 μg·m L-1white wax,and the policosanol aqueous solution which including 6. 25 μg·m L-1tween80 and 12. 5 μg·m L-1policosanol increased significantly the proliferation of HFDPCs comparing with finasteride. [Conclusion]The selected concentration of tween80 is below 6. 25 μg·m L-1in HFDPCs culture when tween80 is used as a solubilizing agent. In the optimistic concentration range of tween80,the white wax and policosanol from white wax could increase the proliferation of HFDPCs remarkably.
