Pb2+与BSA的结合平衡研究
STUDY ON THE BINDING EQUILIBRIUM BETWEEN Pb2+ WITH BSA
【摘要】 在pH 6.3条件下,利用平衡透析法和紫外吸收光谱法研究了Pb2+与BSA的相互作用。用紫外光谱法研究了Pb2+与BSA相互作用的波长为278nm;由Scatchard图得到Pb2+在BSA上的结合位点数为1.6个强结合部位;吸光度随时间的变化反应确定为一级反应;用非线性最小二乘法拟合Bjerrum公式求得逐级稳定常数。
【Abstract】 The binding of Pb2+ to BSA has been studied by equilibrium dialysis and ultra-visible absorption spectrum at p H=6.3, Wavelength about the binding of Pb2+ in BSA by Ultraviolet Visible Spectrophotometer was 278 nm. The Scatchard analysis indicated that there existed 1.6 strong bindings site of Pb2+ in BSA. Reaction of absorbance change over time was a first order reaction. Pb2+-BSA system was determined by non-linear least-square method fitting Bjerrun formula.
