快眼动睡眠剥夺对抑郁模型大鼠海马和下丘脑色氨酸及5-羟色胺水平的影响

【摘要】 目的 探讨快眼动睡眠剥夺对抑郁模型大鼠海马和前额叶色氨酸及5-羟色胺水平的影响。方法 将48只大鼠采用随机数字表法分为4组,正常对照组(A组)、抑郁模型组(B组)、抑郁模型+快眼动睡眠剥夺72 h组(C组)和抑郁模型+大平台对照组(D组),每组12只。A组除行为学实验和定期换垫料外,不给予其他刺激,其它3组连续21 d给予不可预见性刺激,然后对C组大鼠进行72 h快眼动睡眠剥夺。睡眠剥夺结束后采用高效液相-荧光检测法检测大鼠海马和下丘脑部位的5-羟色胺和色氨酸水平。结果 慢性应激和快眼动睡眠剥夺72 h后B组大鼠海马5-羟色胺水平较A组、C组、D组明显降低(P<0.05),5-羟色胺前体物质色氨酸水平较A组明显增高(P<0.05);4组大鼠下丘脑部位.5-羟色胺水平、色氨酸水平比较差异无统计学意义(P>0.05)。结论 快眼动睡眠剥夺可能不是通过改变海马部位5-羟色胺的生物合成,而是通过影响色氨酸的生物利用率而起到快速抗抑郁作用。

【Abstract】 Objective To investigate the influence of rapid eye movement sleep deprivation(REMSD) on hippocampus,TRP,5-HT level in depression rat.Methods 48 rats were selected and divided into 4 groups,normal control group(group A),depression model group(group B),depression model+REMSD 72 h group(group C), depression model + tankcontrol group(group D), 12 cases in each group.Group A was only given behavioral experiment and regular padding change, other 3 groups were given unpredictable stimulation for 21 d, then group C was given 72 h REMSD.After REMSD, hippocampus,TRP,5-HT level of rats were detected by HPLC-FLD.Results Rat hippocampus 5-HT level in group B was obviously lowered compared with that in group A,C and D, 5-HT precusor tryptophan level was obviously increased compared with that in group A,C and D; after 72 h REMSD, the 5-HT in hippocampus showed a reversion, while TRP didn’t.Conclusion REMSD will have rapid antidepressant effects through influence on TRP bioavailability, and may not by changing on biosynthesis of hippocampus 5-HT.
