不同患龋状态双胞胎儿童及其母亲牙菌斑中微生物多样性分析
HOMINGS analysis of plaque microflora in 15 mothers and 30 twins with discordant caries phenotypes
【摘要】 目的:采用人类口腔微生物芯片(HOMINGS-Human Oral Microbe Identification using Next Generation Sequencing)分析拥有不同患龋状态双胞胎及其母亲龈上菌斑中菌群的结构及微生物多样性。方法:牙菌斑样本取自14对双胞胎、1组三胞胎儿童(年龄3~6岁)和15位母亲,共46份样本,分别提取龈上菌斑细菌总DNA,采用HOMINGS测试分析牙菌斑中菌群结构及微生物的多样性。结果:通过高通量测序共获得2293650条有效序列,使用QIIME软件基于97%的相似性聚类OTUs(operational taxonomic unit),得到20431个OTU,经物种注释获得11个门,116个属和139个种水平的物种。在门水平上,母亲组在拟杆菌门和厚壁菌门的丰度显著高于有龋组和无龋组。在属水平上,有龋组和无龋组比较,有龋组在链球菌属、乳杆菌属、放线菌属、嗜血杆菌的丰度要高于无龋组(P<0.05);母亲组的微生物种类较儿童组多,分别在普氏菌属、坦纳氏菌属等10种微生物的相对丰度高于儿童组(P<0.05﹚。结论:母亲组龈上菌斑微生物多样性及优势微生物属水平上丰度显著高于有龋组和无龋组;链球菌属、乳杆菌属、嗜血杆菌属和放线菌属可能为乳牙早期龋(early childhood caries,ECC)的生物标志物。
【Abstract】 Objective: The study was conductedto analyze and compare the oral microbial profiles of the twins with discordant phenotype and their mothers. Methods: Supragingival plaque samples are obtained from 14 paires of twins and a set of triplets with discordant caries phenotypes and 15 mothers,samples are assayed using the Human Oral Microbe Identification using Next Generation Sequencing(HOMINGS). Results: A total of 2,293,650 reads and 20431 OUT revealed 11 phyla,116 genus and 139 species of microflora,the relative abundance of these strains varied dramatically among the children. Comparative analysis between caries group and caries-free group revealed that the frequency of Actinomyces,Lactobacillus,Haemophilus and Streptococcus’ s detection are much higher in caries group(P < 0. 05). The sample from the mother group revealed that the relative abundance of Prevotella,Tannerella were significant higher than caries group(P < 0. 05). Conclusion: The microbial diversity and abundance of mother group was significantly higher than that in caries group and caries-free group. Moreover,the difference of the oral microbiota between caries and caries-free group including Streptococcus,Actinomyces,Lactobacillus and Haemophilus are strongly associated with early childhood caries(ECC) and can be potential biomarkers for dental caries in primary dentition.
