【摘要】 目的:探讨中国汉族精神分裂症患者中5-羟色胺2C受体(5-HTR2C)基因多态性(rs498177)与2型糖尿病共病的关联性。方法:选取长期住院服用氯氮平的精神分裂症伴或不伴2型糖尿病患者(共病组257例和单纯精神分裂症组288例)、单纯2型糖尿病患者(单纯2型糖尿病组264例)及正常人群(正常对照组272名)为研究对象,采用Taqman基因分型技术对5-HTR2C基因的单核苷酸多态性(SNP)(rs498177)进行基因分型,比较各组等位基因、基因型分布频率有无差异以及基因型与代谢指标的相关性。结果:共病组体质量指数(BMI)、腰围、空腹血糖(FPG)及高密度脂蛋白(HDL)水平明显高于单纯精神分裂症组,差异有统计学意义(P均<0.05)。与正常对照组比较,共病组、单纯精神分裂症组及单纯2型糖尿病组5-HTR2C基因型分布上差异有统计学意义(χ~2=22.43,χ~2=12.43,χ~2=15.58;P<0.01或P<0.001);共病组与单纯2型糖尿病组比较,等位基因分布上存在显著统计学意义(χ~2=5.66,P=0.02);精神分裂症患者不同基因型之间BMI、腰围、FPG、TG、HDL差异无统计学意义(F=0.432~2.332,P均>0.05)。结论:在中国汉族精神分裂症患者中5-HTR2C基因多态性(rs498177)与2型糖尿病共病存在关联,其可能是精神分裂症患者患2型糖尿病的易感基因。更多还原

【Abstract】 Objective:To investigate the association of schizophrenia(SCZ) comorbidding type 2 diabetes(T2DM) with the single nucleotide polymorphism(rs498177) of serotonin 2C receptor(5-HTR2C).Method:The Taqman genotyping technique was used to genotype the SNP(rs498177) of 5-HTR2 C in 257long-term hospitalization schizophrenics taking clozapine with T2DM(the comorbidity group),288 long-term hospitalization schizophrenics taking clozapine without T2DM(the SCZ group),264 patients with T2DM(the T2DM group) and 272 normal subjects(the normal control group),and we compare the frequency of allele,genotype and metabolic index of each group.Results:The body mass index(BMI),waist circumference,serum level of fasting blood glucose(FPG) and high density lipoprotein(HDL) in the comorbidity group were significantly higher than the SCZ group(P<0.05).There was no statistically significant difference in age,course of disease,the hospital stay time,clozapine dosage and time and the serum level of TG between these two groups(P>0.05).Compared with the normal control group,the other 3 groups had significant differences in the distribution of genotype of the single nucleotide polymorphism(rs498177) of 5-HTR2C(P<0.05 or P<0.01).There was no statistically significant difference(F=0.432-2.33,P>0.05) in BMI,waist circumference,serum level of FPG,three acyl glycerol and HDL between genotypes of schizophrenics.Conclusion:The single nucleotide polymorphism(rs498177) of 5-HTR2 C in patients with schizophrenia is associated with the comorbidity of T2 DM.It may be a susceptibility gene of this comorbidity in Han Chinese population.更多还原
