ALK2基因敲除小鼠的饲养繁育与基因型鉴定
Breeding,reproducing and type identification for ALK2 gene knock-out mice
【摘要】 目的:为了繁育和鉴定体内ALK2基因被敲除的小鼠,特将从美国引进的ALK2基因敲除小鼠进行饲养繁育,继续保种;以及初步探究激活素受体样激酶2(Alk2)功能缺陷对出生后小鼠在生存能力、繁殖能力、主要组织器官的生长状态等方面的影响。方法:首先用Cre-loxp重组酶系统培育出ALK2条件基因敲除小鼠,通过基因型鉴定筛选出实验组(基因型:Osx-Cre;Alk2fx/fx)和对照组(基因型:Osx-Cre;Alk2fx/+)。结果:成功饲养和繁殖出从美国引进的小鼠,并成功从中筛选出ALK2基因敲除小鼠。结论:采用正确的饲养、繁殖及基因鉴定方法对筛选出基因敲除小鼠及继续保种意义重大;初步探究小鼠ALK2基因敲除后小鼠的变化为进一步深入研究小鼠的组织器官的生长发育方面的变化及机理奠定了基础。
【Abstract】 Objective: Breeding,reproducing,and type identification for ALK2 gene knock-out mice were to preliminary discuss the effect of activated receptor kinase 2( Alk2) defection to survival,reproduction,influence to the growth of tissues and organs. Method: Cre-loxp recombination enzyme system was used to breed ALK2 conditional knockout mice. The mice were divided into experimental group( genotype: Osx-Cre; Alk2 fx/fx) and control group( genotype: Osx-Cre; Alk2 fx/+) at random. Results: The mice imported from the United States were raised and reproduced. ALK2 gene knock-out mice were screened successfully. Conclusion: The proper breeding,reproducing and genetic identification methods are important for screening gene knock-out mice and the preservation of genes; Preliminary investigation into the changes of ALK2 gene knockout mice had laid the foundation for further study on the changes and mechanism of the growth and development of tissue organs.
