SATB2在口腔鳞癌中的表达及其与迁移、侵袭的临床意义研究
Study on the expression of SATB2 andits effct on the migration and invasion of oral squamous cell carcinoma
【摘要】 目的:探讨口腔鳞癌中的核基质蛋白特异AT序列结合蛋白2(SATB2)表达水平的临床意义和对口腔鳞癌生物学行为的影响。方法:收集58例口腔鳞癌标本,检测SATB2的表达情况,分析SATB2表达水平与病理参数之间的关系及与患者总体生存率的关系;用载有SATB2的慢病毒转染HN6细胞,Western blot及实时定量RT-PCR检测转染效果,并通过划痕愈合、Transwell、Invasion实验检测SATB2对HN6细胞生物学行为的影响。结果:统计学结果表明,高表达的SATB2与患者年龄、性别、病理分级无关,但与TNM分期、颈淋巴结转移及临床分期密切相关,与患者的不良预后密切相关;SATB2成功转染HN6细胞系并得到稳定过表达SATB2的Lv-SATB2-HN6细胞;过表达SATB2的HN6细胞迁移、侵袭能力明显高于HN6细胞。结论:SATB2在口腔鳞癌组织及细胞系中表达增高,过表达SATB2能促进肿瘤细胞迁移、侵袭能力。
【Abstract】 Objective: To investigate the expression of nuclear matrix protein-special AT-rich sequence binding protein 2( SATB2)in oral squamous cell carcinoma( OSCC) and its effect on the biological behavior of oral squamous cell carcinoma. Methods: 58 cases of oral squamous cell carcinoma were collected and the expression of SATB2 was detected by immunohistochemical staining. The relationship between SATB2 and the pathological parameters and the overall survival rate was analyzed. Then the HN6 cell lines were transfected with lentivirus containing SATB2 and tested through western blot and RT-PCR as well. Wound healing test,migration tests and invasion tests were preformed for further study of biological roles of SATB2 in HN6 cell lines. Results: The high expression of SATB2 was not associated with age,sex and pathological grade,but was closely related to TNM stage,cervical lymph node metastasis and clinical stage,which was closely related to the poor prognosis. The results of Western blot and PCR showed that SATB2 was successfully transfected into HN6 cell line and Lv-SATB2-HN6 cells stably overexpressing SATB2 were obtained. The HN6 cells overexpressing SATB2 migrated and the invasive ability was significantly higher than that of HN6 cells. Conclusions: The expression of SATB2 in oral squamous cell carcinoma and cell line increased,and overexpression of SATB2 could promote the migration and invasion of tumor cells.
