一种新型两亲性聚乙二醇单甲醚-鹅去氧胆酸聚合物的合成与表征
Synthesis and Characterization of Amphiphilic polymer Poly(ethylene glycol)-Chenodeoxycholic acid
【摘要】 [目的]合成聚乙二醇单甲醚-鹅去氧胆酸聚合物,表征其理化性质。[方法]以聚乙二醇单甲醚和鹅去氧胆酸为原料,先对鹅去氧胆酸进行乙酸化,然后再与聚乙二醇单甲醚进行酯化生成两亲性聚合物聚乙二醇单甲醚-鹅去氧胆酸(mPEG-CDCA)。利用核磁、红外、荧光探针法等手段分析聚合物的结构及理化性质。[结果]mPEG-CDCA被成功合成,且荧光分光光度法测定结果表明其临界胶束浓度(CMC)为17.3×10-3mg/L。[结论]合成的两亲性聚合物有较低的CMC,意味着该聚合物容易形成胶束,在抗肿瘤药物的新剂型中具有良好的研究前景。
【Abstract】 [Objective]The purpose of this paper is to synthesize Amphiphilic polymer and characterize its physicochemical properties.[Methods]The Amphiphilic polymer of Methoxypolyethylene glycols Chenodeoxycholic acid( mPEG-CDCA) was synthesized by the esterified modification on Chenodeoxycholic acid with Methoxypolyethylene glycols. The structural characteristics of copolymers were attested by means of 1H NMR, IR and Fluorescence probe method. [Results]The results of NMR and IR showed that mPEG-CDCA was successfully synthesized and the critical micelle concentration( CMC) was 17. 3 ×10-3 mg/L according to the fluorescence spectrophotometer.[Conclusion]mPEG-CDCA can be successfully synthesized by the synthetic route and method. The results determined by Fluorescence probe method show that Amphiphilic polymer has a smaller critical micelle concentration which means it can be form micelles easily and can be used as a drug carrier. Amphiphilic polymer will have a good prospect in the study of new formulations of antitumor drugs.
