NRF2-SOD2途径参与高海拔暴露引起的肝脏氧化应激及炎性损伤

【摘要】 目的了解慢性高海拔暴露对肝脏氧化应激及炎性反应的影响,初步分析抗氧化应激基因NRF2和SOD2在其中的作用。方法 40只C57BL/6小鼠随机分为正常对照组和高海拔暴露组,每组各20只。其中高海拔暴露组模拟海拔高度6000 m。35 d后测定氧化应激和炎性损伤指标。结果高海拔暴露组小鼠肝脏中抗氧化物GSH-PX(P<0.01)、SOD(P<0.05)明显减少,过氧化物MDA(P<0.05)明显增多,NRF2、SOD2(P<0.01)表达降低。同时高海拔暴露组促炎因子IL-1、IL-6、TNF-α、F4/80(P<0.01)明显升高,抗炎因子IL-10(P<0.01)水平降低。结论慢性高海拔暴露引起小鼠肝脏氧化应激反应及炎性损伤,NRF2-SOD2抗氧化途径可能在这一过程中发挥重要作用。

【Abstract】 Objective To investigate whether chronic high altitude exposure has any effect on liver oxidative stress and inflammatory reactions,and analyze the role of anti-oxidative stress genes NRF2 and SOD2 in this process. Methods Forty C57 BL/6 mice were randomly divided into control and high altitude exposure group. After 35 days of exposure at 6000 meters above sea level,the oxidative stress and inflammatory damage of the liver were determined. Results Such antioxidants as GSH-PX( P< 0.01) and SOD( P<0.05) in the liver of high altitude exposure group decreased significantly,so did NRF2 and SOD2( P<0.01).At the same time,peroxide MDA( P<0.05) increased dramatically,and pro-inflammatory factors IL-6,IL-1,TNF-α,F4/80( P<0.01) in high altitude exposure group increased significantly,while the anti-inflammatory factor IL-10( P<0.01) decreased.Conclusion Chronic high altitude exposure can promote liver oxidative stress and inflammatory injury. NRF2-SOD2 may play a protective role in this process.
