【摘要】 目的探讨黄连素与顺铂联合用药对卵巢癌细胞增殖和侵袭的影响。方法免疫荧光检测不同卵巢癌细胞株对黄连素和顺铂的敏感性;Western blotting检测对不同浓度黄连素和顺铂对抗凋亡蛋白BAG-3的影响;Transwell侵袭实验检测黄连素、顺铂对卵巢癌细胞SKOV3的侵袭能力的影响;平板克隆实验检测黄连素、顺铂对卵巢癌细胞SKOV3的侵袭能力的影响。结果相比A2780细胞株,SKOVE3受黄连素影响明显,25 ng/l浓度的黄连素对SKOV3细胞中BAG-3蛋白的抑制水平最佳;加入黄连素后卵巢癌细胞株SKOV3的侵袭和增殖能力明显降低。结论黄连素与顺铂联合用药可以抑制卵巢癌SKOV3细胞株的增殖和侵袭。更多还原

【Abstract】 Objective To investigate the effect of berberine combined with cisplatin on the proliferation and invasion of ovarian cancer cells.Methods Immunofluorescence was used to detect the sensitivity of different ovarian cancer cell lines to berberine and cisplatin.Western blotting was used to detect the effects of berberine and cisplatin at different doses on antiapoptotic protein BAG-3.Transwell invasion assay was used to detect the effect of cisplatin on the invasive ability of ovarian cancer cell line SKOV3.The effect of berberine on the invasion of ovarian cancer cell line SKOV3 was studied by plate cloning assay. Results Compared to A2780 cells,SKOV3 was significantly affected by berberine.Concentration 25 ng/L of berberine in SKOV3 cells was the most effective for inhibiting the expression of BAG-3.The invasion and proliferation of ovarian cancer cell line SKOV3 were significantly decreased after berberine was added.Conclusion Combination of berberine and cisplatin can inhibit the proliferation and invasion of ovarian cancer SKOV3 cell line.更多还原
