雷帕霉素对肺癌LK2细胞microRNA表达的影响

【摘要】 目的研究药物雷帕霉素作用于非小细胞肺癌LK2细胞后micro RNA表达的变化。方法培养肺癌LK2细胞,研究药物雷帕霉素作用于非小细胞肺癌LK2细胞后micro RNA表达的变化。方法培养肺癌LK2细胞,检测雷帕霉素作用于LK2细胞后其中micro RNA的表达变化,并对表达有显著差异改变的micro RNA进行靶基因预测。结果与对照组相比,雷帕霉素作用于LK2细胞后,9个micro RNAs表达具有高显著差异,其中mi R-320c,mi R-1283,mi R-29b-3p等6个mi RNAs表达显著增强。而mi R-320b,mi R-19b-3p,mi R-181c-6p表达明显降低。结论雷帕霉素作用于肺癌LK2细胞后,可诱导多个micro RNA的表达改变,可以加强部分micro RNAs表达、也可降低多个micro RNAs的表达。

【Abstract】 Objective To study the effects of rapamycin on micro RNA expression of lung cancer LK2 cell in vitro culture. Methods It was objective to culture lung cancer LK2 cells and to study the changes of micro RNA expression after rapamycin treatment in non-small cell lung cancer LK2 cells, target genes were predicted for micro RNA with significant differences in expression. Results The treatment of cells with rapamycin could affect the expression of micro RNAs. Using Illumina Hi Seq TM 2500 mi RNA Sequencing analysis, 9 micro RNAs were found with high expression changes in rapamycin-treated LK2 cells. In these mi RNAs, mi R-320 c, mi R-1283, mi R-29b-3p express higher. mi R-320 b, mi R-19b-3p, mi R-181c-6p express lower. Conclusion After rapamycin act in lung cancer LK2 cells, it can induce multiple micro RNA expression changes, such as enhance and reduce the expression of some micro RNAs.
