【摘要】 目的研究EMP1在喉癌中的作用。方法选取51例喉癌标本及其癌旁组织,构建EMP1过表达载体,转染人喉癌Hep-2细胞系,应用MTT法、克隆形成实验、流式细胞术及Transwell法分别检测细胞的增殖、凋亡及迁移情况,明确EMP1过表达后对喉癌细胞产生的生物学效应。结果 EMP1过表达后,Hep2细胞的增殖能力明显减弱,细胞克隆形成数目平均值明显减少,细胞的凋亡率平均值明显上调,穿过小室滤膜的细胞数平均值明显下调。即喉癌细胞的增殖能力及迁移能力减弱,凋亡增强。结论 EMP1基因在喉癌的发生发展中发挥抑癌基因的作用。更多还原

【Abstract】 Objective To study the function of EMP1 in laryngeal cancer cells.Method 51 cases of laryngeal cancer specimens and its tissue adjacent to carcinoma were selected,EMP1 expression vector was built and transfected into laryngeal Hep-2 cell lines.MTT method,the clone formation test,flow cytometry and Transwell method were used respectively to detect cell proliferation,apoptosis and migration.Results After EMP1 was transfected,Hep2 cells proliferaton capacity was decreased significantly,cells clone formation was significantly reduced,cell apoptosis rate obviously raised,cell number of transwell membrane was significantly decreased.Conclusion EMP1 gene played an inhibition effect on laryngeal cancercells.更多还原
