雷公藤甲素对糖尿病肾病大鼠肾组织iNOS与COX-2表达的影响
Effects of triptolide on expressions of iNOS and COX-2 in renal tissue of diabetic nephropathy rats
【摘要】 目的观察雷公藤甲素对糖尿病肾病大鼠肾组织诱导型一氧化氮合成酶(i NOS)与环氧化酶-2(COX-2)表达的影响。方法选取50只雄性SD大鼠,随机分为正常组(n=10)、模型组(n=20)及雷公藤甲素组(n=20)。模型组与雷公藤甲素组大鼠给予高糖高脂饮食,并以链脲佐菌素腹腔注射建立糖尿病肾病大鼠模型。8周后处死大鼠,检测相应生化指标并计算肾脏指数,免疫组织化学方法和Western blot方法检测i NOS与COX-2的蛋白表达。结果模型组大鼠肾脏指数、血糖、血肌酐和24 h尿蛋白较对照组显著升高,P<0.05;与模型组相比,雷公藤甲素组大鼠的各项指数显著降低,P<0.05。与对照组比较,模型组大鼠肾脏组织i NOS与COX-2的蛋白表达显著升高,P<0.05;与模型组相比,雷公藤甲素组大鼠肾脏组织i NOS与COX-2的蛋白表达显著降低,P<0.05。结论雷公藤甲素能降低糖尿病肾病大鼠肾组织i NOS与COX-2的水平,具有一定的肾保护作用。
【Abstract】 Objective To observe the effects of triptolide on expressions of inducible nitric oxide synthase(i NOS) and cyclooxygenase-2(COX-2) in renal tissue of diabetic nephropathy rats. Methods A total of 50 male SD rats were selected, and randomly divided into control group(n=10), model group(n=20)and triptolide group(n=20). The rats of model group and triptolide group were fed with high-fat diet combined with low-dose streptozotocin intraperitoneally to establish diabetic nephropathy model. After 8 weeks, the rats were sacrificed to detect relative biochemical parameters and renal index, the expressions of i NOS and COX-2 were detected by immunohistochemistry staining and Western blot method. Results Compared with the control group, the levels of blood glucose,serum creatinine and urine protein(24h) were increased remarkably of rats in model group, P<0.05. Compared with the model group, the levels of blood glucose, serum creatinine and urine protein(24h) were increased remarkably of rats in triptolide group, P<0.05. Compared with the control group, the expressions of i NOS and COX-2 were increased remarkably of rats in model group, P<0.05. Compared with the model group, the expressions of i NOS and COX-2 were decreased remarkably of rats in triptolide group, P<0.05. Conclusion Triptolide could provide protection on kidney in diabetic rats by reducing the expressions of i NOS and COX-2.
