PRE-084上调BDNF-TrkB改善PTSD大鼠焦虑样行为
PRE-084 ameliorates anxiety-like behavior via BDNF-Trk B signaling pathway in rat PTSD model
【摘要】 目的探讨腹腔注射Sigma 1受体激动剂PRE-084对创伤后应激障碍(PTSD)大鼠焦虑样行为及额叶BDNF和TrkB表达的影响。方法利用单程长时应激(SPS)制备PTSD大鼠模型,将PTSD大鼠分为4组:vehicle、PRE-084、SPS+vehicle和SPS+PRE-084组;通过旷场实验和高架十字迷宫实验检测各组大鼠焦虑样行为;取材各组大鼠额叶,利用免疫荧光染色和Western Blot检测BDNF和p-TrkB蛋白及m RNA表达情况。结果行为学检测结果显示,腹腔注射PRE-084可以改善PTSD大鼠焦虑样行为;免疫荧光染色和Western Blot结果显示,SPS使大鼠额叶BDNF及p-TrkB蛋白表达水平降低,此作用可被PRE-084逆转。结论腹腔注射Sigma-1受体激动剂PRE-084改善PTSD大鼠焦虑样行为可能与上调BDNF-TrkB信号通路相关。
【Abstract】 Objective To investigate the effect of Sigma-1 receptor agonist(PRE-084) on anxiety like behavior and the expressions of BDNF and TrkB in rats model of posttraumatic stress disorder(PTSD) through intraperitoneal injection. Methods The PTSD model was established by single prolonged stress paradigm(SPS). The animals were divided into 4 groups: vehicle, PRE-084, SPS+ vehicle and SPS+ PRE-084 group. Open-field test, elevated plus-maze test were used to detect the anxiety behavior of rats. The expressions of BDNF and p-TrkB in prefrontal cortex of animals were detected by immunofluorescence staining and Western blot. Results Behavioral test results showed that PRE-084 could ameliorate anxiety symptoms in SPS rats. Immunofluorescence staining and Western Blot showed that the expression levels of BDNF and p-TrkB in prefrontal cortex of SPS rats were improved by PRE-084 injection. Conclusion The ameliorated anxiety behavior of PTSD rats by Sigma-1 receptor agonist(PRE-084) might be related to upregulation of BDNF-TrkB signal molecules.
